Objective:Considering regulatory role of Notch signal pathway in the proliferation and differentiation of keratinocytes, the aim of this study was to observe the expression of Jaggedl, Notchl and NF-κB in Psoriasis patients’ skins by immunohistochemistry, exploring the relationship between signaling pathway abnormalities and evolution of Psoriasis to provide experimental basis of the pathogenesis of psoriasisMethod:According to clinical manifestation and histopathologic results,40patients with acute psoriasis vulgaris were selected in dermatological department of the affiliated hospital of Luzhou medical college during2011-2013. Samples in experimental group were psoriasis plaque of body. Samples in control group were non lesions0.5cm surrounding the lesions from the same patients.In addition,40other patients in dermatological department, department of surgery or burn and plastic surgery were seleted to normal group. All patients were provided the informed consents, and the experiment was approved by the local ethical committee. Observe the expression of Jaggedl, Notch1and NF-κB of skins in each group by immunohistochemistry. Chi-square test and pearson correlation analysis were employed for comparison among groups of all experimental data.Results:1.Analysis of Jaggedl:in the experiment group,40cases of Jaggedl see26cases of positive expression to see12cases of positive expression in the control group40cases,40cases of the normal group to see8cases of positive expression of Jaggedl positive expression rate in the experiment group and control group. The case group and normal group were statistically significant (p <0.017), the control group and normal group showed no statistically significant (p>0.017);2. Analysis of Notchl:in the experiment group,40cases of Notchl see25cases of positive expression to see14cases of positive expression in the control group40cases,40cases of the normal group to see10cases of positive expression of Notchl positive expression rate in the experiment group and control group. The case group and normal group were statistically significant (p <0.017), the control group and normal group showed no statistically significant (p>0.017);3. Analysis of NF-KB:in the experiment group,40cases of NF-KB see25cases of positive expression to see14cases of positive expression in the control group40cases,40cases of the normal group to see10cases of positive expression of NF-K B positive expression rate in the experiment group and control group. The case group and normal group were statistically significant (p <0.05), the control group and normal group showed no statistically significant (p>0.05);4.Jaggedl positive expression of Notchl expression was a significant positive correlation (r=0.643, p<0.05), Jaggedl positive expression and NF-κB positive expression was a significant positive correlation (r=0.649, p <0.05), the Notchl positive expression and positive expression of NF-κB showed a significant positive correlation (r=0.569, p<0.05).Conclusions:1) Jagged1,Notch land NF-κB epidermal expression level was abnormal in the lesions of psoriasis vulgaris throughout the entire course of the disease.2) The enhanced expression of Jaggedl,Notch land NF-κB in the lesion of psoriasis was related to the severity of the disease.3) The expression of Jaggedl,Notch land NF-κB enhancement was closely related to the abnormal cutin cell proliferation, differentiation, which involved in the pathogenic mechanism of psoriasis vulgaris by promoting the activation and adjustment of T cells.4)The positive correlation between the expression of Jaggedl, Notchl, NF-κB,between the two was statistically significant positive expression, from the perspective of Notch signaling pathway put forward the possible pathogenesis with psoriasis vulgaris, and to provide experimental basis. |
