Expression And Significance Of Notch1and Jagged1Proteins In Colorectal Adenocarcinoma Tissue

ObjectiveTo investigate the roles of Notch1and Jagged1protein in the occurrenceand development of colorectal adenocarcinoma(CA).MethodsImmunohistochemical PV-9000method and Western blot were used todetect the expression of Notch1protein and Jagged1protein in78cases of CA（CA group），40cases of colorectal adenoma（adenoma group） and40cases of normal colorectal tissue (5cm far from the CA, normal group)，andtheir relationship with the clinic pathological parameters were analyzed.Results①Notch1protein in colorectal adenocarcinoma positive expression rate of88.5%(69/78) was significantly higher than that of colorectal adenomas, thepositive expression rate of55%(22/40) and colorectal adenocarcinoma of thedistal5cm of normal tissue positive expression rate of42.5%(17/40).Notch1protein in colorectal adenocarcinoma expression and histologic grade,DUCKS stage and lymph node metastasis (P <0.05), and the tissue site, ageand sex (P>0.05).②Jagged1protein in colorectal adenocarcinoma positive expression rate of89.7%(70/78) was significantly higher than that of colorectal adenomas, thepositive expression rate of52.5%(21/40) and colorectal adenocarcinoma of thedistal5cm of normal tissue positive expression rate of47.5%(19/40). Jagged1protein in colorectal adenocarcinoma expression and histologic grade, DUCKS stage and lymph node metastasis (P <0.05), and the tissue site, age and sex (PP>0.05).③Colorectal adenocarcinoma tissue Notch1and Jagged1proteinexpression relations were positively correlated (r=0.407, P <0.01).Conclusions①Colorectal adenocarcinoma group Notch1protein positive rate wassignificantly higher than that of tumor group and normal group (P <0.05).Notch1protein in colorectal adenocarcinoma expression and histologic grade,DUCKS stage and lymph node metastasis (P <0.05), suggesting that Notch1protein in colorectal carcinogenesis process may play an important role.②Colorectal adenocarcinoma group Jagged1protein positive rate wassignificantly higher than that of tumor group and normal group (P <0.05).Jagged1protein in colorectal adenocarcinoma expression and histologic grade,DUCKS stage and lymph node metastasis (P <0.05), suggesting that Jagged1protein in colorectal carcinogenesis process may play an important role.③Colorectal adenocarcinoma tissue Notch1and Jagged1proteinexpression relations were positively correlated (r=0.407, P <0.01). Tip two incolorectal carcinogenesis may play a synergistic effect.
