Object:A conscious heat stroke rat model was established to investigate heat stroke-induced gastrointestinalmotility and changes of serum motilin and gastrin by observing the changes of gastric mucosa glandmorphology and general conditions and Vital-signstã€the serum and gastrointestinal tissue levels ofMTL,GASã€the rate of gastric emptying and the small intestine propulsion in the process of buildingmodel.Methods:1ã€First, building the conscious heat stroke rat model with the use of artificial experimentalenvironment simulation cabin(AEESC) and observing and recording the behavior changes of rat generalconditions and Vital-signs in the process of building model。2ã€the enzyme spectrum index (ALT, AST, BUN, SCR, CK-MB, CK) content were measured withauto bio-chemical analyzer.3ã€The serum and gastrointestinal tissue levels of MTLã€GAS were detected with ELISA.4ã€The gastrointestinal internal marker(blue Sephadex-2000) was tested to determine the rate ofgastric emptying and the small intestine propulsion.5ã€Finally, GAS and MTL expression in gastrointestinal mucosa gland was observed withimmunohistochemical technique,then used the Image-Pro Plus6.0Image processing softwaresemi-quantitative determination the expression quantity of it in gastric tissue。Results:1ã€The control group:activities freely, responsive, free drinking water; the model group:increasedthe amount of water an hour or two after the experiment,activity reductions(4h),supine or gathered,restlessness, the secretion of sweat on abdominal;the above symptoms gradually worsened andunresponsive(16h),fur loosely, the dark yellow hair, Tr(39.98±0.48)℃; sticky secretions around thelips(24h),muscle convulsion, ataxia, slow and deep repiration, listlessness, Tr(41.17±0.84)℃.2ã€Compared with0h group,8h to heat,16h Tr rats increased significantly(P <0.05)whento hot8hã€16h,24h (41to43)℃, MAP value lower (P <0.05), there was significantly reduced at24h (P <0.01);HRlevel increased significantly (P <0.05), the RR significantly speed up (P <0.01),24h group is about0h,8h group of2times value.3〠the enzyme spectrum value(ALTã€ASTã€BUNã€SCRã€CK-MBã€CK)of model rats increasedobviously(P<0.01,P<0.05)by compared with normal control rats.4ã€Compared with normal control rats: the GERS and SIPRS of the rats were significantly lower,while the serum of GAS and MTL levels were higher(P<0.05), peaked at16h then rapidly decreased, thepeak of GAS in gastric tissue moved forward to8h, the longer thermal stimulus time, the lower theconcentration of MTL level in duodenum would be(P<0.05).Conclusion:1ã€A conscious heat stroke rat model was established by a extremely hot and dry environment,the severe heat stroke model complicated after24h.2ã€Pathological change of gastrointestinal mucosal glands tissue damage, gastrointestinalsignificantly motility disorder was found in a conscious heat stroke rat model.3ã€The concentration variation of MTL and GAS in blood can both be an indicator to judgeheat stroke process and to indicate gastrointestinal dysfunction. |
