Objective:To investigate the relationship between SREBP-1c and triglyceride in Huh7cellstransfected recombinant lentiviruses containing PNPLA3gene I148M variant,which isuseful to investigate the pathogenisis of non-alcoholic fatty liver disease.Methods:Huh7cells,transfected the control lentivirus and recombinant lentivirusescontaining PNPLA3gene I148M variant,were classified into2groups: control groupand PNPLA3gene I148M variant group. Change of triglyceride content in2groups wasstained with oil red O. Levels of triglyceride(TG), cholesterol(TC), alanineaminotransferase(ALT) and aspartate aminotransferase(AST) were tested by fullautomatic biochemical analyzer. Expression of SREBP-1c mRNA and SREBP-1cprotein was respectively valued by fluorescence quantitative PCR and Western Blot.Results:Oil red O showed that content of triglyceride in variant group was increasedobviously. Compared with control group, the content of triglyceride in variant groupwas increased significantly ((0.54±0.03) mmol/L vs (0.23±0.02) mmol/L,t=22.58, P<0.01). The content of cholesterol in variant group showed the same changes((0.28±0.03) mmol/L vs (0.13±0.02) mmol/L,t=11.83,P<0.01)).Compared with theircontrol group(92±10)U/L and(279±12)U/L, ALT and AST were increasedobviously(170±10)U/L和(585±12)U/L,P<0.01.Expression of SREBP-1c mRNA wasincreased, contrasted with control group, the difference was statisticallysignificant(P<0.01),and SREBP-1c protein also increased. The level of SREBP-1cwas positively correlated with serum triglyceride in PNPLA3gene I148M variant group(r=0.912,P<0.01).and also was correlated with cholesterol. Conclusion:PNPLA3gene I148M variant increases lipogenesis and SREBP-1c may play a rolein the increase of triglyceride. |
