Objective:A Chinese medical mixture, Wenhua Capsule (WHC), can affect the proliferation, differentiation and apoptosis of human hepatocellular carcinoma cells, leukemia cells and breast cancer cells. WHC has strong cytotoxicity to cancer cells in vitro. Wenhua Capsule can arrest the cell cycle in S phase.5-fluorouracil is an S-phase specific drug and only active during certain cell cycles. The present study was designed to investigate the anti-cancer effects of WHC combined with5-fluorouracil on human breast cancer in vivo.Methods:Nu/Nu mice inoculated with MDA-MB-231breast cancer cells were randomized into the control group,5-fluorouracil group, low-dose WHC (1WHC) group, medium-dose WHC (mWHC) group, high-dose WHC (hWHC) group and combination of mWHC and5-fluorouracil group, respectively. Tumor volume and weight, microvessel density, inhibitive rate of tumor weight, necrosis rate, organ index and body weight of nude mice were estimated.Results:Combining WHC with5-fluorouracil not only enhanced efficacy on inhibition of tumor growth(P<0.05) and microvessel density(P<0.05), but also increased necrosis rate of tumor significantly(P<0.05). The inhibitive rate of tumor weight in comination group of mWHC and5-FU (47.7%)was higher than that in both mWHC group(19.5%) and5-FU group(28.3%). Spleen index in comination group (7.036±1.599) was higher than that in5-fluorouracil group (4.064±1.774)(P<0.01). The increase in body weight of nude mice in combination group(3.52±1.80) was more than that in5-fluorouracil group(1.37±2.06)(P<0.05), and the mice in mWHC group(6.40±2.28) obtained larger body weight than that in hWHC group(3.86±2.42)(P<0.01). Conclusions:1. WHC can significantly inhibit growth of human MDA-MB-231breast cancer.2. Combining WHC with5-fluorouracil can synergistically inhibit growth of human MDA-MB-231breast cancer.3. WHC can significantly inhibit angiogenesis and increase the necrosis rate of human MDA-MB-231breast cancer.4. WHC may improve general condition and immunity of mice with xenografts, and reduce toxicity of chemotherapy.
