Objective: To study the effect of WenHua capsules in the cell proliferation and cell cycle of human breast cancer cell line MCF-7 and T-47D. Method: The experiment was performed in four stages. The first stage, high performance liquid chromatography (HPLC) were carried out to analyze the WenHua capsules constituent and the content of aconitine to contrast the standard preparation. The second stage, the cell line MCF-7 and T-47D were cultured in vitro. To build three observation groups: control group, 75μg/ml WenHua capsules group and 250μg/ml WenHua capsules group. The observation period was seven days. Counting the number of living cells and drawing the curves of growth. To compare the curves of growth with repeated measures analysis of variance. Using inverted phase contrast microscope to observe the alteration of cell morphology. The third stage, to analyze the effect of WenHua capsules in the cell cycle of human breast cancer cell line MCF-7 and T-47D with flow cytometry. The forth stage, to study the effect of WenHua capsules combined 5-Fu in the cell proliferation of human breast cancer cell line MCF-7 and T-47D. To build six observation groups: control group, 75μg/ml WenHua capsules group, 250μg/ml WenHua capsules group, lug/ml 5-Fugroup, 75μg/ml WenHua capsules combined lug/ml 5-Fu group and 250μg/ml WenHua capsules combined lμg/ml 5-Fu group. Drawing the curves of growth and calculating the doubling time. To compare the intensity of cytotoxic via calculating theproportion of died cell. Results: The high performance liquid chromatogram finger printing of WenHua capsules had drawed. The content of aconitine in the WenHua capsules was 1.01 ±0.01 u g. We founded the WenHua capsules had depressant effect to cell proliferation of both cell line. The intensity of depressant effect had positive correlation with drug dose. The observation in cell morphology demonstrated that vacuole-like structures had emerged in the cytoplasm. The quantity of vacuole-like structures kept increasing and mix together. The proportion of cells in S phase were rasised markedly in flow cytometry. In the fourth stage, the depressant effect of WenHua capsules combined 5-Fu was better than the others. But the intensity of depressant effect was independence with the dose of WenHua capsules. Conclusion: WenHua capsules is a complex prescription of pure Chinese drugs pharmaceutics. The manufacture technology is mature and the security is ensured. The cytotoxic action of WenHua capsules is obviously. The WenHua capsules can regulate the cell cycle and induces S phase arrest in human breast cancer cell line MCF-7 and T-47D. If WenHua capsules combined 5-Fu, a cell cycle-specific drug in S phase, the depressant effect would have enhanced. The mechanism of WenHua capsules induces S phase arrest need to be study furtherly.
