Objective: Through the establishment of nonalcoholic fatty liver(NAFLD) rat model, to observe Fangfeng Tongsheng Powder thateffects on non-alcoholic fatty liver, such as serum biochemical levels(AST、ALT、TG、TC)in rats, liver pathology andAQP-9expression,in order to study the effect of Fangfeng Tongsheng Powder treatment ofNAFLD mechanism. Method:60SD male rats(weight in140-200g) wererandomly divided into2groups, after two weeks of adaptive feeding,Normal control group with12rats,which were given normol diet; modelgroup with48rats,which were given high fat diet. After2weeks,48ratswere randomly divided into model control group: Fangfeng TongshengPowder low dose group(6g·kg-1), middle dose group(12g·kg-1),and highdose group(16g·kg-1). Model group and Normal control group withoutany drugs, while were given the same dose of NS. At the end of6,8,10thweek,1-2rats were randomly dissected from the model group todetermine whether successful modeling. After the modeling,drugsintervened two weeks for model control group, then we observedpathological changes in liver tissue and blood biochemical parametersAST、ALT、TG、TC by obtaining venous blood, and all rats weredissected at the end. The liver tissues were fixed with10%formalin, were packaged with paraffin, continuously cut,thickness of slice is5μm, andwas dyed. Furthermore, we detect the expression of AQP-9mRNA in ratliver by RT-PCR method and agarose gel electrophoresis. Result:Innormal control group compared with model control group, serum AST、ALT、TG、TC level, and degree of hepatic steatosis showed significantlyincreased(P<0.01). In model group compared with low, middle, high dosetreatment groups of Fangfeng Tongsheng Powder, the serum AST、ALT、TG、TC level, and degree of hepatic steatosis had statisticallysignificant difference(P<0.05). Low dose group, the expression level ofAQP-9close to the model group, middle dose and high dose werecompared with model group, lower levels of expression than model group(P<0.05), In the high dose of Chinese medicine group was the lowest(P<0.05).Conclusion: Fangfeng Tongsheng Powder can reduce theexpression level of AQP-9in NAFLD rat liver,reduce nonalcoholic fattyliver rats liver fatty degeneration, regulate lipid metabolism disorders,improve liver function to a certain extent, so the AQP-9may be involvedin lipid metabolism in liver, and Fangfeng Tongsheng Powder has thefunction of regulating the expression of AQP-9in rat NAFLD model,which suggests that inhibition of the AQP-9expression perhaps one ofthe mechanisms for the treatment of NAFLD.
