| Objective To compare the regulation mechanism in Lewis tumor-bearing mice treatedwith the Four groups TCM of Yiqi Fuzheng(YQFZ),HuaTan Sanjie(HTSJ),HuoxueHuayu(HXHY),Qingre Jiedu(QRJD) on Common anti-cancer so as to provide thescientific evidences on the clinical efficacy.Methods80SPF grade C57BL/6mice were used and randomly divided into8groupswith the same number of males:blank group,model group,DDP group,Yiqi YQFZgroup,HTSJ group,HXHY group,QRJD group and combined-therapy group.Themodel of Lewis implanted tumor was set up except blank group.The general conditionof mice during experiment was observed,overt behavior, body weights wereexamined.The maximum and minimum diameters of tumor was measured,and volumewas calculated.According the results of volume, portrayed the growth curve.Aftercontinuous medication of14days,the mice were sacrificed.the tumor weight andspleen was measured,tumor inhibitory rate and spleen index was calculated.Thenumber of lung cancer metastasis were observed.HE stain was used to observed tumorpathological changes.FCM was used to detect the changes of cell cycle andapoptosis.IHC was used to detect the changes of tumor tissue VEGF,ES andMMP-2.ELISA was used to detect the changes of peripheral serumAS,ES,VEGF,IL-2,IL-10and IFN-γ in the mice.Results1.The number of tumor lesions on lung metastasis in treatment group hadsignificant difference comparing with Model group group(P<0.05),the DDP groupand combined-therapy group were the least and closest to each other of the number oftumor lesions.No significant changes were demonstrated with treatment group exceptHTSJ group.2.Compared with model group,Tumors’ weight were decreased significantly in thetreating groups(P<0.05).When comparisons were made among each TCM groups,as to combined-therapy group,tumor-suppressor effect was the most significant andtumor inhibition rate were highest.3.In HE pathology work-up,cancer cells had more sparsely distributed in DDP groupand TCM groups than the model group, morphological changes including necrosisarea, karyopyknosis, nuclear fragmentation and nucleus dissolved in the treatinggroups could be observed in the tumors under inverted microscope.4.Compared with the model group, G0/G1phase and apoptosis rate of the treatinggroups were increased (P<0.05),HTSJ group and QRJD group in G0/G1phase ratio ishigher than combined-therapy group(P<0.05),the apoptosis rate in combined-therapygroup was the highest,it did not change obviously in HTSJ group and HXHYgroup.G2/M phase was decreased except YQFZ group and HXHY group(P<0.05),nonetheless,S phase were no significant changes in each group.5.Compared with Model group,the expression of VEGF,MMP-2were decreased andES increased in in treating groups(P<0.05).The VEGF of QRJD group and theMMP-2of combined-therapy group reached the minimum,the ES ofcombined-therapy group reached the maximum among these groups(P<0.05,P<0.01).The DDP group did not have significant changes than other groups had.6.Compared with blank group, the content of VEGF increased significantly in treatinggroups(P<0.05,P<0.01),compared with model group, DDP group,CompatibilityGroups and combined-therapy group were significantly decreased(P<0.01),particularly in DDP group,combined-therapy group and HXHY group;The content ofAS,compared with the blank group, other groups were significantlylower(P<0.01),compared with the model group, DDP group,Compatibility Groupsand combined-therapy group decreased significantly lighter (P<0.01),especially incombined-therapy group and DDP group, QRJD group was most close to the modelgroup;The content of ES,other groups were significantly decreased than those ofblank group (P<0.01),and compared with Model group, the ES levels decreased intreating groups,particularly in DDP group and combined-therapy group.7.After modeling,the spleen indicators and the levels of IL-2and IFN-γ were reducedand the level of IL-10was increased to different extents in each group.The immunity of mice was inhibited.It was found in the comparison among the groups,the spleencoefficients in the individual herbal combined groups and the combined-therapy wereall lower than that in the model group.Concerning to the prevention of IL-2reducing,the results in YQFZ group and HXHY group were better than that in DDPgroup.In term of the prevention of IFN-γ reducing,the result in the combined-therapygroup was significant.It was suggested that the complete formula group achieved thebest effect on IFN-γ.The effect was slightly inferior in the individual combinationgroups.For the prevention of IL-10increasing,the effect was the best in thecombined-therapy group and that was better in HXHY group.Conclusion1.Four compatibility anti-cancer TCM groups had an complementaryinhibitory action on tumorigenesis,its mechanism might be relevant to block the cellcycle on G0/G1phase and promote apoptosis;HTSJ group and QRJD group wasmore prominent through arresting the tumor cell cycle.2.Four compatibility anti-cancer TCM groups could inhibit the expression of VEGFã€MMP-2and enhanced ES level to reduce angiogenesis,QRJD group had moreeffective inhibitory action on VEGF expression,,but the combined-therapy groupwas more effective on decreasing MMP-2and increasing ES.3.Four compatibility anti-cancer TCM groups could inhibit the expression of VEGFand enhanced AS, ES level to reduce angiogenesis.HXHY group and YQFZ grouphad good effect,while combined-therapy group have the best effect.4.Four compatibility anti-cancer TCM groups could protect the spleenfunction,combined-therapy group reached the best effect to regulate IFN-γ andIL-10,the effect was not satisfied with using compatibility groups alone,while usingYQFZ and HXHY group alone had better effects on preventing IL-2fromreducing.The correct compatibility of Four compatibility anti-cancer TCM groupsachieves the coordinative effect inimmuneregulation and anti-tumor to certain extent.... |
