Purpose: Based on the affect of Shugan Liqi Tiaoshen method on expression of proteinkinase A (protein kinase A,PKA)、cAMP response element binding protein (cAMP-responseelement binding protein,CREB) Dysthymia in rat hippocampal of stagnation of liver-QImodel, and to explore the targets and mechanisms on Shugan Liqi Tiaoshen method on theDysthymia of stagnation of liver-QI rat model.Method: The dysthymia of liver-QI model of rats were produced by separation andchronic unpredictable mild stress; Weight test, the consumption of sucrose and open-field testwere used to study the changes of rats behavior; Using the method of chemistry immunityPKA、CREB cell to dye and analyzing the graphics; To observe the ultrastructure of neuronand synapse in the hippocampus of depression rats.Result: The score of weight test、the consumption of sucrose and open-field test of theKeyu shushen Particle array and fluoxetine array are higher than that of the model array’s; Thecontent of PKA、CREB in hippocampal of the curitio array are exceeded than that of themodel array’s; That the ultrastructure of neuron and synapse in the hippocampus of depressionrats suffered pathological changes, and pharmacotherapy can make them improve.Conclusion: Keyu shushen particle can increase the weight and sucrose consumption indysthymia rats, raise the level of movement, vertical movement score, and improvedsymptoms of dysthymia; mechanism Keyu shushen particle may increase PKA and CREB inhippocampus of dysthymia rats thereby affecting the expression of hippocampal neurons,synaptic ultrastructure. |
