A Research On The Effect Of Various Immune Approaches On The Allogeneic Arthroplasty

Purpose:By comparing the different experimental results of various immunological approaches in allogeneic transplant of rabbits’ facet joints,this research is aimed at exploring an effective measure of relieving their immune rejection, accelerating fracture healing and avoiding joint degeneration after the surgery, so as to provide creative ideas for clinical allogeneic arthroplasty.Methods:In this research, 48 adult male rabbits from New Zealand are collected at random, 18 of which are killed a month before the surgery with their third knuckles of the upper limb preserved. The rest are divided into Five Groups as Group A,B,C,D,E, with six rabbits in each group. Direct transplantation is operated on rabbits of Group A, while FK560 are added on those of Group B; Rabbits of Group C are cooled, on which basis Group D are added FK560 for immunity, while rabbits of Group E receive FK560, mycophenolate mofetil and steroids except being cooled.4 weeks later, one rabbit of each experimental group is killed with their specimens observed; 8 weeks later, serum of all the rabbits in each group is distilled to detect their content of Interleukin-Ⅱand Interleukin-Ⅵ; 12 weeks later, imaging inspection is conducted. At the time of 48 weeks, all of the living rabbits in each group are killed for routine HE stained sections to track their recovering from fracture and the condition of cartilage on the articular surface. Finally, the experimental results of each group are compared with their data processed through SPSS16.0statistical software.Results:1) Four weeks after the surgery,except swelling and festering is found in the specimens of three rabbits in Group A to varying degrees, the remaining rabbits in the other four groups healed smoothly.2) Eight weeks after the surgery, the result of observing the Interleukin-Ⅱand Interleukin-Ⅵ in the serum shows that variance analysis indicates the statistical significance in the total difference among all the groups:(F=52.163, P<0.001). According to the comparison between Group B、C and Group A,Group D、E and Group B、C, the statistical result shows P<0.001 while no difference is found in the comparison between Group B and Group A,Group D and Group E with their statistical results respectively showing P=0.150,and P=0.378. The overall results can be stated as follows: A>B、C>D、E.3) The x-ray examination 12 weeks after the operation shows the results of Group B,C,D are evidently more satisfying than those of Group A and B as to the growth of callus and shaping of the cortical bone.4) 48 weeks later, we can get the Wakitani score of the articular cartilage. The score of Group A is far higher than that of any other group,which little difference is found respectively between Group B and C,Group D and E. The overall result show:A>B、C>D、E.Conclusion:Little immune rejection is found in the rabbits in both experimental G roup D and E,whose fracture healed soon and well with low level of furth er joint degeneration, which attained the expected efficacy.Hence the con clusion that pre-operative deep-frozen process combined with immune su ppressants shortly after the operation can not only effectively relieve the r ejection of the receptor and promote the healing between the donated bon e and its original one but also effectively prevent its further joint degenera tion so that the function of the donated knuckle can be better preserved to the extreme extent in the body of the receptor.
