Objective: The characteristics of histology and immunology were investigated after tendon allografts had been transplanted. Methods: NewZealand donor rabbits' Achilles's tendon auto grafts and fresh Achilles's tendon allografts after cut off were immediately transplanted into corresponding NewZealand acceptor rabbits. Donor rabbits' Achilles's tendon after cut off which were frozen to -26℃ and kept 10 day as ordinary-frozen allgraft providers, donor rabbits' Achilles's tendon after cut off which were slowly frozen to -80℃ and kept 10 day as deep-frozen allgraft providers, after 10 day these two group providers were transplanted into corresponding acceptors. After transplanted four weeks, acceptors Achilles's tendon grafts were harvested and immediately fixed by glutaraldehyde and dyed with HE method, then observed under experimenters naked eyes and transmission electronic microscope and microscope. At the same time experimenter measured function of cellular immunity through lymphocyte mortality test and lymphocyte proliferating test and phagocytizing rate of celiac macrophages. Result: 1) under scope: (1)Deep-frozen group: Tenocyte proliferating was activity, its quantities were numerous, intracellular apparatus was up growth, a lot of rebirth and thin collagen fibril could be seen, and a little inflammatory cell could be seen, collagen fibril arranged in regular and tight, rebirth capillary could be seen at anywhere, its characteristic was similar to tendon auto graft transplantation; (2)Ordinary-frozen group: a few tenocyte could be seen, intracellular apparatus function was inferior, butstill could be seen a little rebirth collagen fibril, inflammatory cell quantities were more than deep-frozen group, a little rebirth capillary could be seen, collagen fibril arranged in regular and tight but curve collagen fibril looked as wave could be seen in local. (3)Fresh allograft group: could not nearly see tenocyte, could not see any new collagen fibril at all, thick collagen fibril arranged in disorder and loose, inflammatory cells could be seen anywhere. 2) Lymphocyte mortality and lymphocyte proliferating rate and phagocytizing rate of macrophages in the deep-frozen group was lower than ordinary-frozen group (P<0.05), ordinary-frozen group was lower than the fresh allograft group (P< 0.05),there were no statistical difference between the auto graft group and the deep-frozen group (P>0.05). Conclusion: Immune rejective action in ordinary-frozen group was more severity than the deep-frozen group and the auto graft group. Immune rejective action in the deep-frozen group was quite lower, its characteristics of histology and immunology after transplantation was similar to the auto graft group.
