Objective:Selecting Primary(Ovariectomized) and secondary(injected of dexamethasone) osteoporotic SD rats. Measured the rats’ femoral BMD by dual-energy X-ray absorption(DXA) and observed HE staining in non-quantitative indicators of bone cells and bone matrix structure. How the endogenous H2 S generated in the blood and bone was dynamic observed, to clarify inhibition of endogenous H2 S generation and generation-related osteoporosis.Methods : 6-month-old adult SD rats were 48 in total, of which non-pregnant female SD rats were 36 and male SD rats were 12, both body weighted 240 ± 20 g. SD rats were randomly divided into four groups. Group A : matched group, male and female rats were 12 in total, intramuscular saline 2.5mg / Kg, 3 times a week.Group B: sham operation group, non-pregnant female SD rats 12(only around ovary removal of adipose tissue, without removal of ovaries). Group C: surgical group, non-pregnant female SD rats 12(ovariectomized). Group D: hormone group, male and female rats were 12 in total, intramuscular dexamethasone 2.5mg / kg, 3 times a week.A, B, C, D four groups of rats were detected whole body BMD and femoral BMD at the preoperative and 12 weeks, to judge whether or not a successful model built. After the success of the model built at 2,4,6,8 weeks each group sacrificed three rats and detected serum and bone tissue of endogenous H2 S content by isolated femoral BMD and HE staining electron microscopy morphology.Results:1. There were no difference in body BMD and femoral BMD(P> 0.05) of A, B rats before or 12 weeks after surgery; There were differences in body BMD and femoral BMD(P <0.05) of C, D rats before or 12 weeks after surgery; At the same time point after surgery,there were significant differences between C, D groups of rats and A,B rats in body BMD and femoral BMD(P <0.05).2.Comparing A,B,C,D groups at 2,4,6,8 weeks after the successful construction of osteoporotic models. There is no significant difference between C and D groups(P> 0.05) or A and B groups(P>0.05); while group C compared with A, B(P <0.05), group D compared with A, B(P <0.05).3. Scanning optical microscope observation group at 2,4,6,8 weeks after the successful construction of osteoporotic models. Group B compared with C,D: the matched group(group A) and group B were normal bone cells, uniform distribution, the number of bone cells did not significantly change over time, bone matrix did not appear obvious breakage or fracture phenomenon, the smaller the gap, the smaller the number of voids, no dispersion phenomenon or uneven distribution. Group D and C stayed normal shape and size of bone cells the number of osteoblasts operation group over time more normal bone cell shape and size over time, but a sharp reduction in the number of osteoblasts, the bone matrix breakage or fault clearance clear, cortical bone loosely arranged, more empty and scattered, sparse, uneven distribution.4. At 2,4,6,8 weeks after the successful construction of osteoporotic models,there were no significant differences between every groups(P> 0.05), indicating the existence of endogenous H2 S in the blood system was too wide, little difference. There were also no significant difference in bone tissue at the same time points 2 and 4 weeks(P>0.5).But at 6 and 8 weeks time point, there were significant differences between group C, D and group A, B(P<0.01). Conclusions: Primary and secondary osteoporosis SD rat bone changes correlated with endogenous H2 S.
