Objective:The study is designed to monitor the effects of JGP on bone metabolim of the ovariectomized rats.Then we can assess the effect and explore the mechanism of JGP on the ovariectomized rats.Therefore to provide a scientific,reasonable and theoretical guidance and treatment for clinical prophylaxis and treatment of PMO.Method:We make rat osteoporosis model by ablating bilateral ovaryies on 50 rats.10 rats were sham operation as normal control group.The experimental rats were divided to five groups.They are model group,estrogen group,high dose group of JGP,middle dose group of JGP and low dose group of JGP.After treatment,we compared the value of bone mineral density(BMD)between model group and sham group,to determine whether the model of PMO had been establish successfully or not.Effects of estrogen and different dosage levels of JGP on the PMO rats are assessed by measuring BMD, Bone tissue morphometry,biochemical indicater and made ANOVA analysis.Result:1.After ovaries are excised for 3 months,the BMD in rats of modle group were significantly lower than that of sham group(p<0.01).It indicates that the animal model of PMO has been establish successfully.2.Compared with modle group,JGP has the better effect of preventing and treating PMO.The BMD of all JGP group increased obviously.(P<0.05-0.01).3Compared with modle group,the level of serum E2 increases obviously in all JGP group(P<0.05-0.01).But the level of serum calcium,ALP and BGP in middledose group of JGP and low dose group of JGP decrease obviously(P<0.01),otherwise the content of IGF- I in blood serum increase obviously (P<0.05).4 Bone histomorphology is analysised by computerized image analysis system.The result shows that the percentage of average bone trabecula areas(TS%) and width of average bone trabecula(WAT) improve obviously in all JGP group(P<0.05-0.01).The quantity of bone scrobiculus unit area of sham group is significantly higher than that of modle group(P<0.01),but there is no statistic difference between JGP group and modle group.Conclusion:JGP has the effect of preventing and curing PMO,its effect is equal to that of E2.The effect of JGP on PMO is partly attributed to the raising of serum E2 level and suppressing the high-turnover status of bone.
