【Objective】 To investigate the expression level of FAP-1protein in cholangiocarcinoma cancer tissues and adjacent tissues and explore the correlation of FAP-1, Bax and Bcl-2 with cell apoptosis, and providing clinical reference value for the prognosis, and provides a new way for the future research on the prevention and treatment of cholangiocarcinoma cancer.【Methods】 All tissues were obtained in Hunan Provincial People’s Hospital from June 2008 to June 2014, all samples were not treated with radiotherapy and chemotherapy before operation. The samples contained cancer tissues and adjacent tissues, all the tissues were confirmed by pathology. Immunohistochemical(S-P) method was used for detecting the expression level of FAP-1, Bcl-2 and Bax protein in 63 cases of paraffin carcinoma tissues and 42 cases of adjacent tissues in paraffin. And the expression level of FAP-1, Bax and Bcl-2 protein of 36 cases of cancer tissues and 32 cases of adjacent tissues were detected by Western blot. The expression levels of FAP-1、Bcl-2 and Bax in cancer tissues and adjacent tissues, and the relationship of FAP-1、Bcl-2 and Bax were statistically analyzed. Furthermore, the relationship between FAP-1 and clinicopathological features was analyzed, too.【Results】(1) The expression levels of FAP-1 and Bcl-2 in cancer tissues were significantly higher than adjacent tissues and Bax was significantly down-regulated in cancer tissues.By the Pearson correlation analysis, FAP-1 protein and Bcl-2 protein in cholangiocarcinoma were positively correlated(R=0.724,P<0.05), and FAP-1 protein and Bax protein in cholangiocarcinoma were negatively correlated(R=-0.683,P<0.05). The positive expression rates of FAP-1 in cholangiocarcinoma cancer tissues and normal adjacent tissues were 82.5%(52/63) and 9.5%(4/42) and the positive expression rates of FAP-1 in cholangiocarcinoma cancer tissues was significantly higher than normal adjacent tissues(P<0.01). The positive expression rates of Bcl-2 in cholangiocarcinoma cancer tissues and normal adjacent tissues were 77.8%(49/63) and 4.8%(2/42) and the positive expression rates of Bcl-2 in cholangiocarcinoma cancer tissues was significantly higher than normal adjacent tissues(P<0.01). The positive expression rates of Bax in cholangiocarcinoma cancer tissues and normal adjacent tissues were 25.4%(16/63) and 97.6%(41/42) and the positive expression rates of Bcl-2 in cholangiocarcinoma cancer tissues was significantly lower than normal adjacent tissues(P<0.01). The positive expression rates of FAP-1 and bcl-2 were increased with later clinical pathology, lower histological degree and patients with node metastasis. And the positive expression rate of Bax was decreased with later clinical pathology, lower histological degree and patients with node metastasis.(2) The up-regulation of FAP-1 protein in cholangiocarcinoma cancer was significantly correlated with tumor growth, and the expression of FAP-1 was related with clinical pathology, histological degree and lymph node metastasis(P<0.05).(3) Bcl-2 protein was up-regulated in the cancer tissues which were positive expression of FAP-1 and Bax protein was down-regulated in the cancer tissues which were positive expression of FAP-1. By the Pearson correlation analysis, FAP-1 protein and Bcl-2 protein in cholangiocarcinoma were positively correlated(R=0.783,P<0.05), and FAP-1 protein and Bax protein in cholangiocarcinoma were negatively correlated(R=-0.679,P<0.05).【Conclusion】(1) The expression of FAP-1 and Bcl-2 were significantly up-regulated in cholangiocarcinoma cancer tissues and Bax was down-expressed.(2) The up-expression of FAP-1 in cholangiocarcinoma cancer tissues was related with clinical pathology, histological degree and lymph node metastasis(3) FAP-1 protein and Bcl-2 protein in cholangiocarcinoma were positively correlated,and FAP-1 protein and Bax protein in cholangiocarcinoma were negatively correlated.(4) The positive expression of FAP-1 protein may be through down-regulating the expression of Bax and up-regulating the Bcl-2 expression, due to decrease the ratio of Bax/ Bcl-2, and induce apoptosis inhibiting excessive. And leading to the occurrence and development of cholangiocarcinoma cancer.
