Backgroud Gastric ulcer is one of the most common gastric disease in China, about 3.96% in adult, among them 5%~10% change into cancer. The incidence of gastric ulcer is strongly associated with infection of Helicobacter pylori (Hp), which induced gastric ulcer interacted with gastric, pepsin, invasion of gastric mucosa and genetical factors. More and more attention were paid on non-coding RNA with regulation of gene transcription recently; However, research on lncRNA is still at early stage, the role of lncRNA in regulation of transcription in gastric ulcer have not been reported.Purpose This study aims to investigate role of lncRNA in Hp associated with gastric ulcer and its molecular mechanism, the results will provide new therapeutic strategies to treat with gastric ulcer.Methods Outpatients and inpatients with gastric ulcer from January 2014 to October 2014 in department of Endoscopy were collected and analyzed, then divided into Hp-positive and Hp-negative groups. Expression of lncRNA-MEG3 in gastric mucosa were detected with RT-PCR, then evaluating the relationship between gastric inflammation and expression of lncRNA. The regulation of lncRNA and MUC5AC on Hp associated with gastric ulcer were detected with Immunohistochemistry (IHC), RT-PCR, Western blot.Results ① Expression of MUC5AC in Hp positive were higher than that in Hp negative gastric tissue by immunohistochemistry method. ②Nonparametric test results showed that it was positive correlation between low expression of lncRNA mRNA and Hp-positive. Among 40 patients with gastric ulcer, of which 30 cases with Hp-positive gastric ulcer lncRNA-MEG3 mRNA low expression in 22 cases (73.3%) and high expression of 8 patients (26.7%), of which 10 cases with Hp-negative lncRNA-MEG3 mRNA low expression in 3 case (30%) and high expression in 7 cases (70%); ③ High mRNA expression of lncRNA-MEG3 in normal gastric mucosal cell GES-1, verus low mRNA expression of lncRNA-MEG3 in Hp-positive gastric cancer cell AGS and MKN28; ④ Compared with placebo, mRNA expression of lncRNA-MEG3 were significantly lower with the treatment of Hp in GES-1, P<0.05.Conclusion Hp promote the occurrence of gastric ulcer by mediated binding with down-regulation of lncRNA-MEG3 and up-regulation expression of MUC5AC.
