Part1 Determination of the serum IL-12 level in the patients with tuberculosis of bone and joint and healthy individuals in Guangxi Zhuang populationObjective: To explore the diference of serum IL-12 level between the patients with tuberculosis of bone and joint and healthy individuals in Guangxi Zhuang population, as well as its clinical significance. Methods: The concentration of serum IL-12 was measured by ELISA in 150 patients with tuberculosis of bone and joint(case group) and 165 helahty individuals(controls group) in Guangxi Zhuang population. The ESR level and CRP level were measured by routine method. Analysis the correlation of IL-12 and ESR levels in the case group, also anlysis the correlation of IL-12 and CRP levels in the case group. To investigate the relationship of serum IL-12 level and thetuberculosis. Result:(1) The level of serum IL-12 in the case group was significantly lower than that in the controls group(P<0.01). Furthermore the levels of ESR and CRP were higher in the case group than that in the controls group(P<0.01).(2) Levels of ESR and CRP were not significantly correlated with the concentraiton of serum IL-12 level(P>0.05). Conclusion: The serum IL-12 level of in the case group was lower than that in healthy individuals.IL-12 may play a role for immunoregulation in the pathogenesis of tuberculosis.Part 2 Studies on the association between IL-12 single nucleotide polymorphisms and susceptibility to tuberculosis of bone and joint in Guangxi Zhuang populationObjective:(1) To explore the differences about genotypes and alleles frequency of IL-12 A gene single nucleotide polymorphism rs568408 G/A and IL-12 B gene single nucleotide polymorphism rs3212227 A/C between Guangxi Zhuang population and other races in different regions.(2) To explore the relationship between the SNPs polymorphisms and the susceptibility to tuberculosis of bone and joint in Guangxi Zhuang population,the relationship between genotypes of SNPs and the the serum IL-12 level is also evaluated. Methods:(1) The IL-12 A gene rs568408 G/A and IL-12 B rs3212227 A/C polymorphisms were detected by polymerase chain reaction-single base extension technique and direct DNA sequencing in 150 patients of tuberculosis of bone and joint(case group) and 165 healthyindividuals(control group) in Guangxi Zhuang population.(2) the genotypes and alleles frequency of SNPs in control group were compared with the other four populations(Hap Map-HCB, Hap Map-JPT, Hap Map-YRI, Hap Map-TSI)from Hap Map database.(3) The genotypes and alleles frequency of SNPs in case group and control group were analyzed by Statistic and the relationship between the genotypes and alleles frequency of SNPs and susceptibility to tuberculosis is evaluated.(4) Analysis the association between genotypes of 2SNPs and serum IL12 concentration which detected before. Results:(1)There were IL-12 A gene rs568408 G/A and IL-12 B gene rs3212227 A/C polymorphisms in case group and control group.(2) The genotypes and alleles frequency of rs568408 in Guangxi Zhuang population were not significantly differenct with Hap Map-HCB, Hap Map-JPT and Hap Map-TSI(P>0.05), but were significantly different with Hap Map-YRI(P<0.01), the genotypes and alleles frequency of rs3212227 in Guangxi Zhuang population were not significantly differenct with Hap Map-HCB and Hap Map-JPT(P>0.05), but were significantly different with Hap Map-YRI and Hap Map-TSI(P<0.01).(3) The genotypes and alleles frequency of SNPs were not significantly different between case group and control group(P>0.05).(4) The comparison on serum IL-12 level of rs568408 site of GG wild type and GA+AA mutant type in case group have no significat differencers(P>0.05), the comparison on serum IL-12 level of rs3212227 site of AA wild type and AC+CC mutant type in case group have no significat differencers(P>0.05). Conclusion:(1) There are significant differences in the genotypes and alleles frequency of IL-12 A gene rs568408 G/A and IL-12 B gene rs3212227 A/C between Guangxi Zhuang population and other ethnic populations, and this variation might contribute for a variety of clinicalmanifestation and morbidity of some IL-12 related diseases.(2)Polymorphisms of IL-12 A gene rs568408 G/A and IL-12 B gene rs3212227A/C have no effect on susceptibility to tuberculosis of bone and joint in Guangxi Zhuang population, the single nucleotide polymorphisms of IL-12 A gene rs568408 G/A and IL-12 B gene rs3212227 A/C may have no relationship with serum IL-12 level. |
