Objective: To study the association between single nucleotide polymorphism(SNP)of MASP1 gene rs3774268,rs17040 locus and TAGLN2 gene rs2252815,rs2789422 locus and genetic susceptibility to hepatocellular caracinoma(HCC)in families which are come from Guangxi Fusui Zhuang population.Methods: We selected twenty families with high incidence of hepatocellular carcinoma(including 20 patients and 59 healthy relatives)and 10 normal control families(40 persons)from the Zhuang nationality in Fusui,Guangxi.The DNA,of tissue or blood samples was extracted and the genotypes of rs3774268 and rs17040 loci of MASP1 gene and rs2252815 and rs2789422 loci of TAGLN2 gene in DNA samples were detected by time-of-flight mass spectrometry(TOF-MS).The goodness of fit x~2 test was used to test the Hardy-Weinberg equilibrium of each group of genotypes to determine whether the subjects were representative of the population.The relationship between the genotypes and alleles of four loci of MASP1 and TAGLN2 and the incidence of hepatocellular carcinoma was analyzed by unconditional logistic regression equation.Results:1.Genotypic test results of MASP1 gene rs3774268 and rs17040 loci(1)there were three genotypes of AA,GG and GA at rs3774268 locus of MASP1 gene.The frequency of AA genotype in HCC group,non-HCC group and normal control group was 5.0%,12.5% and 0.0%,respectively.The frequency of GG genotype in HCC group,non-HCC group and normal control group was 65.0%,52.5% and 87.5%,respectively.The frequency of GA genotype in HCC group,non-HCC group and normal control group was 30.0%,39.0% and 12.5%,respectively.The actual frequency and predicted frequency of genotype AA,GG and GA were in good agreement with each other in the three groups(P > 0 05).(2)the rs17040 locus of MASP1 gene has two genotypes: CC and CT.The frequency of genotypic CC in HCC group,non-HCC group and normal control group was 80.0%,81.0% and 95.0%,respectively.The frequency of genotypic CT in HCC group,non-HCC group and normal control group was 20.0%,19.0%and 5.0%,respectively.The actual frequency and predicted frequency of genotype CC and CT were in good agreement with each other in group C(P >0.05).2.The relationship between MASP1 gene rs3774268,rs17040 single nucleotide polymorphisms and susceptibility to hepatocellular carcinoma(1)There are two allele-types at rs3774268: G and A,three genotypes: AA,GG and GA.In the non-hepatoma group,individuals who with allele A were1.55 times(95% CI: 0.65 to 3.72,P=0.323)more likely to develop hepatocellular carcinoma than G individuals;the risk of hepatocellular carcinoma in individuals who with genotype GA was 18.20 times(95% CI: 0.56 to 595.71,P=0.103)more than that of GG individuals;the risk of hepatocellular carcinoma in individuals who with genotype AA was 0.05 times(95% CI: 0.00 to 5.75,P = 0.219)more than that of GG individuals.In the normal control group,individuals with genotype A had a 3.75 times higher risk of developing liver cancer than individuals with genotype G(95% CI 1.14 to 12.35,P < 0.003).The risk of hepatocellular carcinoma in individuals with GA and AA genotypes could not be calculated.(2)There are were alleles in the rs17040 locus: C and T,two genotypes:CC and CT.In the non-hepatocarcinoma group,the risk of hepatocellular carcinoma in individuals with allele T were 0.94 times(95% CI: 0.28 to 3.15,P=0.924)more than C individuals;the risk of hepatocellular carcinoma in individuals with genotype CT was 0.36 times(95% CI: 0.03 to 4.67,P = 0.559)than that of CC individuals.In the control group,the risk of hepatocellular carcinoma in individuals with allele T were 0.23 times(95% CI: 0.04 to 1.32,P= 0.099)more likely to develop hepatocellular carcinoma than C individuals;individuals with genotype CT had hepatocellular carcinoma The risk cannot be calculated.3.Genotypic test results of TAGLN2 gene rs2252815 and rs2789422 loci(1)there were three genotypes of CC,CT and TT at rs2252815 locus.The frequency of genotypic CC in HCC group,non-HCC group and normal control group was 5.3%,16.9% and 20.5%,respectively.The distribution frequency of genotypic CT in HCC group,non-HCC group and normal control group was52.6% and 37.3%,respectively.The distribution frequency of genotypic TT in HCC group,non-HCC group and normal control group was 42.1%,45.8% and 41.0%,respectively.The actual frequency and predicted frequency of 41.0%CC,CT and TT in the three groups were in good agreement with each other.(2)there were three genotypes of CC,CT and TT at rs2789422 locus.The frequency of genotypic CC in HCC group,non-HCC group and normal control group was 47.4%,59.3% and 70.0%,respectively.The distribution frequency of genotypic CT in HCC group,non-HCC group and normal control group was52.6% and 39.0%,respectively.The distribution frequency of genotypic TT in HCC group,non-HCC group and normal control group was 0.0%,1.7% and5.0%,respectively.The actual frequency and predicted frequency of genotype CC,CT and TT were good in the three groups(P > 0 05).4.Relationship between s2252815 and rs2789422 single nucleotide polymorphisms of TAGLN2 gene and susceptibility to hepatocellular carcinoma(1)There were two alleles at rs2252815: C and T,three genotypes: CC,CT and TT.In the non-hepatocarcinoma group,individuals with allele C were 0.84times(95% CI: 0.38 to 1.82,P=0.651)more likely to develop hepatocellular carcinoma than T individuals,and the risk of hepatocellular carcinoma in individuals with genotype CT was 0.75 times(95% CI: 0.09 to 6.15,P = 0.786)than that of TT individuals,and the risk of hepatocellular carcinoma in individuals with genotype CC could not be calculated.In the normal control group,individuals with allele C were 1.43 times(95% CI: 0.63 to 3.25,P=0.392)more likely to develop hepatocellular carcinoma than T individuals,and individuals with genotype CT had hepatocellular carcinoma The risk was 0.45times(95% CI: 0.10 to 2.05,P=0.303)that of TT individuals,and individuals with genotype CC were 2.06 times(95% CI: 0.13 to 34.10,P=0.612)more likely to develop hepatocellular carcinoma than TT individuals.(2)There were two alleles at rs2789422: C and T,three genotypes: CC,CT and TT.In the non-hepatocarcinoma group,individuals with allele C were 1.33times(95% CI: 0.57 to 3.10,P=0.511)more likely to develop hepatocellular carcinoma than T individuals,and the risk of hepatocellular carcinoma in individuals with genotype CT was 0.49 times(95% CI: 0.05 to 5.34,P=0.559)than that of CC individuals,and the risk of hepatocellular carcinoma in individuals with genotype TT could not be calculated;compared with the liver cancer group,the normal control group contained individuals with locus C were at 0.59 times(95% CI: 0.24 to 1.50,P=0.269)more likely to develop hepatocellular carcinoma than those with T,and individuals with genotype CT were at risk for hepatocellular carcinoma were 0.53 times(95% CI: 0.11 to 2.49,P = 0.417)more than that of CC individuals,the risk of hepatocellular carcinoma in individuals with genotype TT could not be calculated.Conclusion:1.The distribution of various genotypes of MASP1 gene rs3774268,rs17040 locus and TAGLN2 gene rs2252815 and rs2789422 locus in the Fusong Zhuang population of Guangxi is consistent with Hardy-Weinberg equilibrium rule and has a representative population.2.There is a significant correlation between the single nucleotide polymorphisms of MASP1 gene rs3774268 and the genetic susceptibility of hepatocellular carcinoma with high incidence in Guangxi Fusong Zhuang population.Allele A is a risk factor for hepatocellular carcinoma in Fusong Zhuang population in Guangxi.3.There is no correlation between the MASP1 gene rs17040 locus single nucleotide polymorphisms and the genetic susceptibility of hepatocellular carcinoma with high incidence in the Fusong Zhuang population in Guangxi.4.The single nucleotide polymorphisms of TAGLN2 gene rs2252815 and rs2789422 were not associated with the genetic susceptibility of hepatocellular carcinoma with high incidence in Guangxi Fusong Zhuang population.
