Background:Gastric cancer is one of most frequent malignant tumors in the world,especially in the east asia. It is the major cause of morbidity and cancer death. With the development of modern medicine, various diagnosis and treatment methods occur。 Owing to the CT and gastroscope’s development,we diagnosis more and more early stage gastric cancer. However, Clinically its incidence is increasing gradually and gastroscope as a screening test is difficult to acquire its mass appeal. Many patients have developed into advanced gastric cancer when they are diagnosed, invasion and metastasis has occurred. They lost the best chance of surgery, the effect of radiotherapy and chemotherapy is always limited. The current study has found that the incidence of tumor invasion and metastasis is related to the development of cancer stem cells more or less. In order to unlock the mysteries of gastric cancer and to improve the rate of gastric cancer diagnosis and survival rate, we need to research gastric cancer stem cell and its related mechanisms.Objective:To research the distribution and expression level of CD44 and Prpc.To analysis the relationship between the two genes with the aspects of gastric cancer’s proliferation, invasion, metastasis and prognosis.And to further explore the molecular mechanisms and related signal transduction pathways.Method:1. Gastric cancer patients were collected and followed up. To observe the distribution and expression level of CD44 and Prpc in in normal gastric mucosa and gastric tissue by immunofluorescence and immunohistochemical.And to analyze the relationship between the two proteins with gastric cancer clinical pathology.2. To screen the high expression level of CD44 and Prpc in the gastric cancer cell lines by Real-time quantitative PCR and Western blot. We construct the prpc sh-RNA plasmids and the corresponding expression plasmids. Then to analyze the impact of silence Prpc and high expression of EMT-related proteins by RNA interference technology. We observe the impact of prpc on the colony-forming by plate clone formation assay.3. We collect fresh blood of Gastric cancer patients and healthy people. We use flow cytometry to analyse of the percentage of cases of double positive cells and single-positive cells.Result:1.Immunohistochemistry and immunofluorescence tissue show:CD44 and Prpc expression rate in gastric cancer was significantly higher than in normal gastric mucosa. The expression of CD44 and Prpc with gastric cancer invasion, lymph node metastasis and tumor stage is positively correlated.2. The level of the expression of CD44 and Prpc in gastric cancer cell lines MGC 823 is relatively high. Silencing the expression of Prpc MGC823 can reduce invasion and metastasis of gastric cancer cells, clonogenic capacity and reduce the expression of N-cadheren.3. Patients with gastric cancer, the circulation of CD44 and Prpc double positive cells and single-positive cells are relatively higher than that of healthy population.Conclusion:1. The expression of CD44 associated with Prpc can be used as a biomarker of tumor invasion, metastasis and malignant indicators, and may become a prognostic indicator in the future.2. silencing prpc gene expression can Inhibit gastric cancer cell invasion and metastasis, proliferation.Simultaneous the expression of Prpc may also affect the expression of EMT-related proteins.3.The level of CD44 and Prpc double positive tumor cells in the circulatory system is significantly higher thanthat of the healthy population, there is likely to be a chance through the blood to diagnosis cancer.4. CD44 and Prpc play an important role in tumor progression and metastasis, especially Prpc may be as a potential tumor marker, it may become a special cancer target for diagnosis and treatment in the future.
