Objective To develop a HPLC method for the simultaneous determination of Scopolin,Scopoleti and Chlorogenic acid in Obtuseleaf Erycibe Stem herbs.To study and establish the fingerprint chromatogram for the identification of Obtuseleaf Erycibe Stem.Methods A HPLC analysis was carried out on a Hypersil ODS(4.6 mm×200mm,5μm) with the mobile phase consisting of acetonitrile(A)-0.1% aqueous phosphoric acid solution(B) in gradient mode.The detection wavelength was set at 347 nm and the column temperature was kept at 30℃.The flow rate was 1.0m L·min-1.Results There was a good linear relationship between the concentration and peak area in the range of 0.399-31.920μg·m L-1,0.360-28.800μg·m L-1,0.412-32.960 μg·m L-1 for scopolin,scopoletin and chlorogenic acid(r=0.9999,n=6),the average recoveries of scopolin,scopoletin and chlorogenic acid were 99.97%(RSD=1.28%),99.49%(RSD=0.60%),99.92%(RSD=0.99%),respectively.Throug h the 12 batches of different origin Obtuseleaf Erycibe Stem herbs determination to establish a Obtuseleaf Erycibe Stem herbs Fingerprint mode,the calibration of the 10 peaks,and through the "TCM fingerprint evaluation software," to calculate the 10 batches of medicinal similarity in 0.9,2 batches of medicinal similarity in 0.9.Conclusions This method is simple and reliable,and can simultaneously determinetion of scopolin,scopoletin and chlorogenic acid content of three active ingredients in Obtuseleaf Erycibe Stem,can be used for medicinal Obtuseleaf Erycibe Stem quality control.
