Objective: This study detected the expression of GPR124 in gastric cancer tissues, analyzed its clinical and pathological significance, and provided a valuable reference to the early diagnosis and molecular targeted therapy of the gastric cancer.Methods: 20 cases of fresh specimen of gastric cancer, paracarcinoma and normal tissues were collected respectively. The protein and total RNA was extracted and the expression of GPR124 was detected by Western blot and RT-PCR. Meanwhile, 99 cases of paraffin specimens of different differentiated adenocarcinoma, atypical hyperplasia and normal gastric tissues were collected respectively. The positive rates of the expression of GPR124 in different gastric tissues were detected by immunohistochemistry staining. Then, the expression and the significance of the GPR124 were analyzed.Results:1.Western blot showed that the levels of the expression of GPR124 protein in gastric cancer were much higher than in paracarcinoma and normal tissues(P<0.05), and the levels of the expression of GPR124 protein in paracarcinoma tissues were higher than in normal tissues(P<0.05).2.RT-PCR showed that the levels of the expression of GPR124 RNA in gastric cancer were much higher than in paracarcinoma and normal tissues( P<0.05), and the levels of the expression of GPR124 RNA in paracarcinoma tissues were higher than in normal tissues(P<0.05).3.Immunohistochemistry staining showed that GPR124 protein was expressed on cell membrane and the positive expression rates of the gastric cancer, atypical hyperplasia and normal tissues were 77.78%(49/63)、30.77%(4/13)、4.35%(1/23)respectively. The positive rates of the expression of GPR124 protein in gastric cancer were much higher than in atypical hyperplasia and normal tissues(P<0.05),and the positive rates of the expression of GPR124 protein in atypical hyperplasia were higher than in normal tissues(P<0.05).Meanwhile, the positive rates of the expression of GPR124 protein in well differentiated, moderately differentiated and poorly differentiated gastric cancer tissues were 33.33%(4/12)、75.00%(15/20)、96.77%(30/31),there were significant difference in these three groups(P<0.05).The positive rates of the expression of GPR124 protein in stage III~IV gastric cancer were much higher than stage I~II,and in gastric cancer with lymphatic metastasis were much higher than in gastric cancer with no lymphatic metastasis.Conclusions: 1.The expression of the GPR124 protein was up-regulated in the gastric cancer tissues.2.The regulation of the GPR124 protein was related to the different differentiation of the adenocarcinoma,clinicopathological staging and lymphatic metastasis.
