ObjectiveTo observe the effect of Compound phyllanthus Urinsria Ⅱ (CPUⅡ) in the rat model of liver cancer induced by DEN on regulating abnormal network that contain IL-6 centered signaling pathway and microRNA related to inflammation-associated cancer, explore the deeper level of its anti cancer mechanism.MethodAll experimental Wistarmale rats were randomly divided into five groups, blank group, high-and low-dose CPU II groups, tegafur group. Rat model of liver cancer was induced by DEN for eighteen weeks, and medicine intervention was made at the same time according to the experimental design for eighteen weeks. Taken once every 6 weeks,8 rats in each group of rats were observed and, generally, the indexes of liver function changes and liver nodule formation. With routine HE staining and Edmondson classification, Rat serum IL-6 use radioimmunoassay detection; the change of gene mRNA expression in liver tissue were detected by real time fluorescent quantitative PCR and the change of gene protein expression in liver tissue were detected by Western blot in the signal pathway of IL-6 and MicroRNA regulatory network.ResultsIn the end of eighteenth weeks, compared with that in the model group, the numbers of liver carcinoma nodules in rats in high-and low-dose CPU II groups, were significantly reduced (P<0.05), the level of IL-6, ALT and AST in the serum of high-and low-dose CPU II groups were all decreased too (P<0.05), but serum level of ALB increased (P<0.05). After 18 weeks, the expression level of MicroRNA let-7a in liver tissues increased comparing with that of model group (P<0.05), but the mRNA expression level of NF- kappa B/P65, IL-6, Lin28, STAT3, RAS and c-myc decreased(P<0.05). The expression level of miR-21 and miR-181b-5p in all groups of CPU II were decreased in liver tissues (P<0.05), and the protein expression level of STAT3, p-STAT3 and CYLD were also obviously reduced(P<0.05).ConelusionCompound phylla CPUⅡ can downregulate let-7a gene expression, the up regulation of NF-kappa B/P65 expression, thus the inhibition of IL-6 expression, blocking the activation of the IL-6 signaling pathway, then inhibited the occurrence and development of hepatocellular carcinoma induced by DEN, in a word, CPUⅡ can prevent and treat liver cancer.
