The Study Of Changes Of AMPK And RAGE Expression In Hepatocelluar Carcinoma Bearing Mice Treated With Metformin

Objective:To explore antitumor effect of metformin on hepatocelluar carcinoma by making Related intervention on tumor nude mice,detect AMPK expression in traditional view And AGEs/RAGE axis expression to study the posssible relationship between both?Methods:1.Using the hepatocellular carcinoma strain HepG2 cultured in vitro subcustaneously inject to the left armpit of the BALB/C nude mice constructed the tumour-bearing nude mice constructed the tumor-bearing model.Randomized to blankgroup,BSA group,AGEs group,metformin group and the AGEs/metformin combined intervention group.2.measuring weight change of nude nice, fasting glucose, the long diameter(a),short diameter(b)of the tumor size at regular intervals.3.four weeks after intervention, taking sample of blood,let it stay for the night,1000 rpm, 20 minutes to save supernatant in- 80 ? condition,after nude mice were executed,take sample of hepatocellular carcinoma tissue, normal liver tissue, tumor tissue adjacent and save them in- 80 ?condition.4.by applying the method of ELISA and immunohistochemical to detect expression of AMPK and RAGE in each tissue sample.Results:1.metformin can inhibit tumor growth, can reduce a tumor-burdened rat body weight(P < 0.05), has no hypoglycemic effect on the tumor-burdened rat's fasting blood glucose(P>0.05).2.AGEs ELISA can detect out low AGEs level in the serum of nude mice which recive tail vein AGEs injection, both injected with metformin and AGEs nude mice serum,other groups' outcome is lower than the lower limit value.3. using immunohistochemical method measure the expression of AMPK, RAGE,can discover that metformin can rise AMPK expression, reduce the expression ofRAGE.AGEs also has effect on RAGE.Conclusions:Metformin surely has antitumuor effect on HCC tumor-bearing nude mice model,can reduce weight of nude mice,has no significant effect on fasting glucose.which may activate AMPK,lower RAGE to give play to the role of anti-tumor,long time AGEs intervention maybe play a role in enhacing AMPK expression and decreasing RAGE expression. The study of AMPK and RAGE,it may provide a new train of thought for metformin anti-tumor mechanism.
