ObjectiveTo determine the effects of XZCs on hepatic steatosis in Sprague-DawIey rats and elucidate the relevant mechanisms. To investigate the effects of drug-containing serum with Xiaozhi Decoction on the differentiation and mechanism in 3T3-L1 Adipocytes.MethodsHepatic steatosis was induced in the rats with high-fat, high-cholesterol feeds. Intracellular lipid droplets were detected with oil red 0 staining, and liver injury was examined by hematoxylin and eosin staining to calculate the NAFLD activity scores (NASs; hepatic steatosis, inflammation, and ballooning degenera-tion of liver cells). Biochemical parameters, including total trigly-ceride (TG), total cholesterol (TC), aspartate transaminase (AST), gamma glutamyl transpeptidase (GGT), alanine transaminase (ALT), malondial-dehyde (MDA), superoxide dismutase (SOD), glucose, free fatty acids(FFAs), leptin, and adiponectin, were measured wi th commercial kits. Sterol regulatory element-binding protein-lc (SRRBPlc), tumor necrosis factora (TNFa) and peroxisome proliferator-activated receptor α (PPAR a), expression was alyzed by western blots and/or real-time reverse transcription-polymerase chain reaction. Use different concentration of drug-containing serum with Riaozhi Decoction in 3T3 L1 Adipocytes, cell proliferation was determined by eck-8, cell different)tiation was determined by Oil Red 0 staining and spectrophotometry, PPARα mRNA expression and liver X receptor (LXRα) mRNA expression was examined by reverse transcription polymerase chain reaction (RT-PCR).ResultsModerate and high doses of XZCs reduced lipid droplet accumulation; hepatic TG and TC contents; GGT, AST, ALT, adiponectin, and MDA levels; and NASs, and increased SOD and leptin activities (P<0.05 or P<0.01). These doses enhanced (P<0.01) PPAR α expression in a concentration-dependent manner and reduced (P<0.05 or P<0.01) SREBPlc and TNFα expression. There was no significant affect on the proliferation of 10% drug-containing serum with Xiaozhi Decoction in 3T3-L1 Adipocytes (P>0.05), however, it could significantly inhibit 3T3-L1 preadipocytes into mature fat cells (P<0.05 or P<0.01), and there is a dose-dependent relationship. Compared with the control group,10% drug-containing serum with Xiaozhi Decoction could significantly reduced PPARα mRNA expression and liver X receptor mRNA expression.ConelusionXZCs exert a preventive effect against hepatic steatosis, and the potential underlying mechanism might be the alteration of PPARα, SREBPlc, and TNFa expression.10% drug-containing serum with Xiaozhi Decoction could significantly inhibit 3T3-L1 preadipocytes into mature fat cells, the mechanism may be related to it could significantly reduce PPARα mRNA expression and liver X receptor mRNA expression.
