| Objective:Investigation the role and significance of inflammatory and endoplasmic reticulum stress in the pathogenesis of NAFLD by observing the expression of inflammasome-associated protein TLR4/NF-κB and SREBP-1c in liver tissues of non-alcoholic fatty liver disease.Methods:NAFLD patients confirmed by Liver biopsy were taken as the research object,and compared with normal liver tissues of patients with hepatic hemangioma.To observe the expression of TLR4 、 NF-κB and SREBP-1c in liver tissues of patients with NAFLD and their relationship with clinicopathological features by immunohistochemical staining,semi quantitative analysis.Results:1.Inflammation of the liver and steatosis Indexes between NAFLD group and normal control group: ALT(122.00(65.00,425.00)vs38.30±7.33,z=4.400 p < 0.001)、 AST(222.10±127.30vs30.40±3.98,t=6.728 p < 0.001)、 GGT(307.55±167.47vs23.70±8.19,t=7.562 p<0.001)、Degree of liver steatosis(F)(2.65±0.23 vs 0.5(0.0,1.0),z=7.515 p<0.01)、Inflammation grade(G)(1.50±1.05 vs0),the difference between the two groups was statistically significant.2.Expression of TLR4 in liver tissue and its relationship with inflammation and steatosis The positive expression rate of TLR4 in liver tissues specimens of patients with NAFLD was 98%,a few positive expression in normal liver tissues.The difference between the two groups was statistically significant(x2=11.273 p=0.001).The expression level of TLR4 in liver tissues was positively correlated with the degree of liver steatosis(rs =0.817,p = 0.001)and inflammation grade(rs=0.601,p=0.005)of NAFLD patients.3.Expression of NF-κB in liver tissue and its relationship with inflammation and steatosis2.2 The positive expression rate of NF-κB in the liver tissues of NAFLD patients was100%.No positive expression in normal liver tissues or a small number in the liver cytoplasm.The difference between the two groups was statistically significant(x2=12.000,p=0.001).The expression level of NF-κB in liver tissues of NAFLD was positively correlated with the degree of liver steatosis of NAFLD(rs =0.756,p< 0.001)and inflammation grade(rs =0.798,p<0.001).4.Expression of SREBP-1c in liver tissue and its relationship with inflammation and steatosis The positive expression rate of SREBP1 c in the liver cytoplasm of NAFLD patients was 100%,a few positive expression in normal liver tissues,the difference between the two groups was statistically significant(x2=15.000,p < 0.001).The expression level of SREBP1 c in liver tissues of NAFLD was positively correlated with the degree of liver steatosis of NAFLD(rs =0.601,p=0.005)and inflammation grade(rs=0.704,p = 0.008).Conclusions:1.Inflammasome-associated protein TLR4/NFκB was highly positive in liver tissues of NAFLD patients,and its expression level was positively correlated with the patient’s inflammatory and degree of liver steatosis.It is suggested that activation of TLR4 receptor and activation of NF-κB may play a role in the pathogenesis of NAFLD.2.SREBP-1c was highly positive in liver tissues of NAFLD patients,and it was positively correlated with the degree of liver steatosis and inflammation grade.It is suggested that endoplasmic reticulum stress also play a role in the pathogenesis of NAFLD. |
