Objective:To observe the different effects of X-ray and Heavy Ion Beams on the expression of vascular endothelial growth factor (VEGF) and integrin αVβ33 in Tca8113 cell line of human tongue squamous. To evaluate how different radiation affects the tumor angiogenesis and hematogenous transfer of tumor cell, and to provide a new basis for cancer therapy by Heavy Ion.Methods:Human tongue squamous carcinoma Tca8113 cell line was kindly provided by Wuhan University. The cells were cultivated for the following experiments, which was dived into the control group, X-ray irradiation group and heavy ion beam irradiation group. X-ray and Heavy Ion Beams were used for the radiation and the cells were irradiated by different time(12h,24h and 48h) and doses of these kinds of radiations. The indirect immuno fluorescent experiment was used to estimate the expression of VEGF and integrin αVβ33 of the cells. Faxitron RX-650 type X-ray apparatus with the light energy of 50 kVp was used for the experiment, and the dose rate was 0.5Gy/min. The HIRFL terminal with the light energy of 250 MeV and LET of 75 KeV was used for the irradiation, and the dose rate was about 1 Gy/min. The radiation dose of control group was O. and the doses of experiment groups were 1Gy,2Gy, and 4Gy.Results:1. Compared with control group, there was no difference on the expression of VEGF in X-ray group at different times and in different doses; however, with the dose of 2Gy or 4 Gy, there was an obvious reduction in Heavy Ion Beams group at the 24th hour after radiation (p<0.05). While with the dose of 1Gy,2Gy or 4 Gy there was also an obvious reduction in Heavy Ion Beams group at the 48th hour after radiation (p<0.05).2. Compared with control group, there was obvious increment on the expression of αVβ3 in X-ray group with the doses of 4Gy and at the 24th hour after radiation. While, with the dose of 1Gy or 2Gy there was also an obvious increment in X-ray group at the 48th hour after radiation. In the Heavy Ion Beams group, the expression of αVβ3 was increased with the condition of 12th hour and 4Gy, while the expression of αVβ3 in the Heavy Ion Beams group was significant reduced at the condition of 12h,48h(4Gy) (p<0.05).Conclusion:Compared with X-ray, Heavy Ion Beams is a better way to inhibit the expression of VEGF of Tca8113 cell line in vitro. However, there was no difference on the expression of VEGF in X-ray group at different times and in different doses. There was a obvious increment on the expression of αVβ3 in X-ray group with the doses of 4Gy and at the 24th hour after radiation and 48h (1Gy、 2Gy). On the contrary, the expression of αVβ3 in the Heavy Ion Beams group was significant reduced at the condition of 12h,48h(4Gy).This may indicate that Heavy Ion Beam is a better way to inhibit the cell invasion, which may provide a new method for the radiotherapy of Human Tongue Squamous Cell Carcinoma.
