Objective:1.To explore the different effects of X-ray and Heavy Ion Beams on the expression of chemokine receptor CXCR4 and RhoC mRNA in Tca8113 cell line.2. To explore the different effects of X-ray and Heavy Ion Beams on the expression of CXCR4 and RhoC protein in Tca8113 cell line.3. To evaluate how different radiation affects the tumor invasion through the expression of CXCR4 and RhoC, and to provide a new basis for cancer therapy.Methods:Human tongue squamous carcinoma Tca8113 cell line was kindly provided by Wuhan University and stored by Institute of Modern Physics, Chinese Academy of Sciences. The cells were cultivated for the following experiments. X-ray and Heavy Ion Beams were used for the radiation and the cells were irradiated by different doses of these kinds of radiations. The real-time PCR experiment was used to estimate the expression of CXCR4 and RhoC mRNA of the cells. And the Western blot experiment was used to evaluate the expression of CXCR4 and RhoC protein. The HIRFL terminal with the light energy of 250 MeV and LET of 75 KeV was used for the irradiation, and the dose rate was about 1 Gy/min. Faxition43885D type X-ray apparatus with the light energy of 50 kVp was also used for the experiment, and the dose rate was 0.5Gy/min. The radiation dose of control group was 0, and the doses of experiment groups were 1Gy,2Gy, and 4Gy, under the different doses group the cells were gathered at the 4th hour,8th hour,12th hour, 24th hour after radiation separately, depending on the experiment design.Results:1.Compared with control group, there was a minimum of the expression of CXCR4 in X-ray group at the 8th hour after radiation, and a peak at 12th hour after radiation (p<0.05), but the trend was not significant in the 2Gy group (p>0.05); however, with the same dose there was an obvious reduction in Heavy Ion Beams group (p<0.05). Meanwhile, the difference of the expression of CXCR4 was more obvious than that in X-ray group (p<0.01).2.Compared with control group, the relative expression levels of RhoC mRNA in X-ray and HI group were both inhibited (P<0.05). And there was basically the same trend with the different radiation doses of 1Gy,2Gy, and 4Gy. In the X-ray group, the expression of target gene was increased with the dose, while the expression in Heavy Ion Beams group was inhibited with the increasing dose. And after radiation, the expression of RhoC mRNA in the Heavy Ion Beams group was significantly reduced compared with the blank control group (P<0.05).3.Western blot resluts showed that at the 12th hour after radiation, the expression of RhoC was increased along with the increasing dose, while the change of CXCR4 was unobvious. And at the 24th hour after radiation, the expression of RhoC was decreased along with the ingcreasing dose, and the trend of CXCR4 was same but unobvious. The resluts were mainly in line with the real-time PCR experiment.Conclusion:Compared with X-ray, Heavy Ion Beams is a better way to inhibit the expression of CXCR4 mRNA of Tca8113 cell line in vitro. Although the expression of RhoC mRNA was inhibited in both X-ray and Heavy Ion Beam groups, inhibition of RhoC in heavy ion beams group was superior to that in X-ray group. This may indicate that Heavy Ion Beam is a better way to inhibit the cell invasion, which may provide a new method for the radiotherapy of Human Tongue Squamous Cell Carcinoma.
