The Impact Of Guiqi Decoction For Laxativation On Constipation Model Mice Defecation And The Vasoactive Intestinal Peptide

Objective:This experiment was intended to apply of Guiqi decoction for laxativation to constipation model mice, by observing the mice general behavior, the first time of discharging black defecate and black grain numbers within six hours, weighing their wet weight and dry weight,calculating water content, detecting colon vasoactive intestinal peptide content, to explore the action mechanism of Guiqi decoction for laxativation treatment of functional constipation in children. Methods:To prepare sixty SPF Kunming mice, half male and half female, which were randomly divided into six groups,as follows: normal group(A), distilled water group(B), model group(C), Ma Ren pills group(D), Guiqi decoction for laxativation with small dose group(E)and high-dose group(F). Except group A, group B was given distilled water to fill the stomach,the other four groups were given compound diphenoxylate suspension liquid lavage, one time a day, continuous ten days, before and after building model, observed their general behavior and defecate character. The 11 th day to 17 th day, group A would not be given any processing, group B and group C were given distilled water to fill the stomach, the other three groups were given the corresponding drug lavage, one time a day, seven days continuous lavage. Except group A,the mice of other five groups,in the 11 th day, 14 th day and 17 th day after feeding corresponding drug lavage,twenty minutes later ink would be used to fill the stomach, and then recorded every mouse’s first time of discharging black defecate within six hours, grain number, wet weight, dry weight and water content three times; The 18 th days,after filling the stomach,all mice were broken neck death,which were used to test colon vasoactive intestinal peptide content according ELISA kit construction. Results:1. The first day before building model, six groups mice had good mental state,sensitived in response and fur light, their defecation appearance looked round,big shape and high gloss. After ten days building model,the general condition of group B had no obvious change,but the model groups spirits were drooping and laged in response, they were easy to gather together and reduced food intake,their defecation appearance looked hard,small shape and low gloss. 2.After seven days corresponding processing, compared with group B, group C’s was obviously prolonged in first time of discharging black defecate within six hours, and also their black defecate number were decreased, which had statistical significance(P<0.01). Compared with group C, group D and group F were significantly shortened in the first time of discharging black defecate within six hours, and their black defecate number were increased, which had significant differences(P<0.01). Compared with group B, group C’s black defecate wet weight and water content had significantly reduced(P<0.01); Compared with group C, group D, group E and group F,those three groups mice ’s black defecate wet weight and water content increased obviously, which had significant differences(P<0.01). 3.Compared with group A, group B ’s colon VIP content had no obvious difference(P>0.05); Compared with group B,group C ’s colon VIP content was significantly decreased, which had statistical significance(P < 0.01); Compared with group C, group D, group E and group F,those three groups mice’s colon VIP content was significantly increased, which had statistical significance(P<0.01). Conclusion:1.The Guiqi decoction for laxativation can improve constipation model mice’s defecate condition.2.The Guiqi decoction for laxativation can shorten the constipation model mice’s first time of discharging black defecate within six hours,increase their black defecate number,wet weight and water content within six hours.3.The Guiqi decoction for laxativation can increase constipation model mice’s colon vasoactive intestinal peptide content.
