| Objective:To observe changes of vasoactive intestinal peptide-immunoreactive (VIP-IR) neurons in colonic enteric nervous system (ENS) after treatment with traditional Chinese medition Qing Yi Decoction in rat severe acute pancreatitis.Methods:Thirty female Sprague-Dawley rats (200g~230g) were randomly divided into three groups:a sham operated group (n=10), a SAP group (n=10), a SAP group treated with Qing Yi Decoction (QYD group)(n=10). SAP model was induced by retrograde cholangiopancreatic duct injection of5%sodium taurocholate. Rats in Sham operated group and SAP group were introgastrically given saline1h,2h and3h after operation. Rats in QYD group were introgastrically given Qing Yi Decoction1h,2h and3h after SAP induced. Intestinal transit and Pathological damage of pancreas and colonic mucosa was studied. Double-immunolabeling for Hu-IR neurons and VIP-IR neurons was employed for the whole-mont preperation of colonic submucosal ganglia and colonic my enteric ganglia in ENS. Results:In sham operated rats, Hu-IR neurons were25.13±7.90per myenteric ganglion and12.48±5.01per submucosal ganglion. The percentage of VIP-IR neurons per myenteric ganglion was lower than per submucosal ganglion (12.94±4.82%vs67.63±8.08%). The percentages of VIP-IR neurons in colonic myenteric ganglia (44.28±7.08%) and submucosal ganglia(67.63±8.08%) in the SAP group were significantly higher compared with the sham operated group (12.94±4.82%in myenteric ganglia and40.76±8.96%in submucosal ganglia, P<0.05). The percentage of VIP-IR neurons of QYD group was between sham operated group and SAP group:29.14±5.92%in colonic myenteric ganglia and56.98±7.10%in submucosal ganglia (P<0.05). The scores of pathological lesion grade of pancreas were3.1±0.4in sham operated group,12.6±1.8in SAP group,5.9±0.6in QYD group, respectively. Small intestinal transit index were53.32±4.67%in sham operated group,17.21±3.98%in SAP group,29.41±1.29%in QYD group, respectively. The small intestinal transit index in the SAP group was lower compared with the sham operated group(P<0.05). Conclusions:Improvement of SAP early gastrointestinal dysfunction by traditional Chinese medition Qing Yi Decoction may be involve in plasticity of VIP-IR neurons in the enteric nervous system. |
