ObjectiveTo detect in the expression of multi-resistant associated protein 1 (multi-drug to hold relevant proteinl, MRP1) and tongue cancer drug resistance gene (TCRP 1) in human Oral squamous cell carcinomas (Oral squamous cell carcinoma OSCC), and explore the relationship between their expression and multidrug resistance(MDR),reveal the correlation between the expression of TCRP1 and MRP1, investigate the relationship between their expression and clinical characters.MethodsRandomly collected 30 patients with oral squamous cell carcinomas cancerous tissue under aseptic conditions, cancer types are squamous cell carcinomas, each cancer tissue were divided into two,one used for the original generation of cultured cells, single cell suspension and planting in 96-well plates, the cells for drug sensitive test, and then determined by MTT method were used to detect cell proliferation activity of cancer cells resistant index calculation; another part of the fixed, paraffin embedding, make-up the paraffin sections and immunohistochemical detection TCRP1 and MRP1 expression in these organizations, and then the results are analysis semi-quantitative integral method, statistical analysis of drug susceptibility in vitro experimental results with TCRP1 and correlation of MRP 1 expression.Results(1) Primitive cells of oral squamous cell carcinomas in vitro medicine sensitive experiment results show that:26 cases of 30 cases of oral squamous carcinoma tissue to obtain valid data, of which 8 cases of drug-resistant,18 cases of sensitive, sensitive rate was 69.2%, is almost the same with the clinical use of cisplatin sensitive rate.(2) Oral squamous carcinoma tissues immunohistochemical results show:in 30 cases of oral squamous carcinoma specimens, TCRP1 and MRP1 expression are mainly located in cytoplasm and nucleus.Including TCRP1 expression of 22 cases were negative or weakly positive; 8 cases of positive or strong positive expression. The expression of MRP1 in 21 cases were negative or weakly positive; 9 cases positive or strong positive expression. TCRP1 positive rate was 66.7%, MRP1 positive rate was 73.3%.In medicine sensitive experiment in different degree of sensitivity to cisplatin, TCRP1 and the expression of MRP1 quantity is different also, basic present positive correlation.Using the exact probability statistical method to calculate the expression of MRP1 and TCRP1 in sensitive and resistant group difference, found the susceptibility and resistance groups TCRP1 expression differences are statistically significant (P< 0.05);and the expression of MRP1 in the group of drug susceptibility and resistance difference is statistically significant (P< 0.05). Results suggest drug susceptibility in vitro experiment and immunohistochemical results have consistency; both expressed in drug-resistant group was obviously higher than that of sensitive group.TCRP1 and the expression of MRP 1 results are linear correlation analysis, the results showed that the correlation coefficient r= 0.742 (P< 0.05), can be concluded that the expression is closely related to.In the resistant group,the linear correlation analysis, the expression of MRP1 and TCRP1 has nothing with the patients age (P> 0.05); By t test, the expression of TCRP1 and MRP1 were no TNM staging, gender difference (P> 0.05).ConclusionThe TCRP1 and MRP1 resistance rate of cisplatin in vitro drug are consistent with cisplatin clinical use rate. The high expression of TCRP1 and MRP1 in oral squamous cell carcinoma has associated with drug resistance of OSCC. TCRP1 and MRP1 expression was positively correlated, possible synergies in oral squamous cancer drug resistance. TCRP1 and MRP1 expression in drug resistance groups are closely related.
