Abstract/Summary:
Objective:To investigate the inhibitive effect of MRP1-siRNA on human tongue squamous cell carcinoma multidrug resistance.Methods:One siRNA-MRP1 was designed, synthesized and transfected into human tongue squamous cell carcinoma cell line Tca8113/CDDP.Observe the dynamic transfection through microscope and confirm the transfection effeciency.Expression level of MRP1 mRNA was detected through RT-PCR.MRP1 protein expression was assayed with immunocytochemistry.MTT was used for observing the cytotoxic efect of Cisplatin(CDDP) after MRP1-siRNA transfected.Results:After MRP1-siRNA transfected to Tca8113/CDDP cell, expression of MRP1 mRNA obviously decreased,the expression level of MRP1 mRNA decreased61.3%. Trough 36 h after transfection, positive rate of MRP1 protein expression in MRP1 siRNA group was obviously decreased compared with the control group.The inhibition of CDDP on Tca8113/CDDP cell growth in MRP1 siRNA group was obviously increased compared with the control group and the inhibitive effect was strengthen gradually along with the time.Conclusions:MRP1-siRNA can obviously inhibit the multidrug resistance of human tongue squamous cell carcinoma cell line Tca8113/CDDP.
