Objective:To investigate The effect of cyclooxygenase-2Inhibitor NS-398onproliferation, apoptosis and cox-2expression of human gastric cancer SGC-7901cell,and explore its possible mechanism.Methods: methyl thiazol tetrazolium(MTT) method was used to detectantiproliferative ratio of NS-398on human gastric cancer SGC-7901cell;Immunocytochemical detection of gastric cancer cells COX-2protein expression;immunosorbent assay(ELISA) method was used to detect the release levels of PGE2of NS-398on human gastric cancer SGC-7901cell; flow cytometry to analyze thegastric cancer cell apoptosis.Results:NS-398inhibited the proliferation of SGC-7901cells in a time-andconcentration-dependent manner (P<0.05).The expression of COX-2significantlydecreased after the administration of NS-398(P<0.05).NS-398significantly inhibitedthe release of PGE2in SGC-7901cells in a dose-dependent manner (P<0.05).Theapoptosis of SGC-7901cells increased after the administration of NS-398for48hours, and the apoptosis increased more and more with the concentration ofNS-398(P<0.05).Conclusion:NS398can inhibit proliferation of human gastric cancer SGC-7901cell and induce its apoptosis in vitro,which may contributed to the COX-2dependentpathway. |
