Objective: Wnt/β-catenin signaling pathway can influence and regulate the self-renewal and differentiation of stem cells,play an important role in the process of cell proliferation,differentiation,exercise and apoptosis,its abnormal activation or expression is closely related with the occurrence and development of tumor.Wnt1 is a member of Wnt gene family,is the start factor of Wnt signaling pathway,some study confirmed that Wnt1 can promote the occurrence of multiple tumors. β-catenin, a Mr 92,000 protein that also functions in cadherin-based epithelial cell adhesion, is a key regulator of Wnt signaling.,the high expression of β-catenin is associated with poor prognosis of tumor.Lgr5(leucine-rich repeat-containing g-protein coupled receptor 5) aslo known as GPR49,which belongs to the G protein coupled receptor family,is a newly target gent of Wnt pathway,but also a kind of potential colon cancer stem cell markers,It has been demonstrated to be involved in different human cancer entities,including hepatocellular carcinoma,basal cell carcinoma,colorectal cancer,ovarian cancer,esophageal cancer and so on. Chronic superficial gastritis(chronic superficial gastritis, CSG) is a chronic inflammation of gastric mucosa caused by a variety of incentives, belonging toone of the common diseases,digestive system. Its not given timely treatment or improper treatment can easily progress to chronic atrophic gastritis and cancer.in this study,we constructed a chronic superficial gastritis rat model and to observated the changes of expression of Wnt1,β-catenin and Lgr5 in gastric tissue,We want to find the relationship with gastric tumorigenesis and progression and to early detective and prevent the development of gastric cancer.Methods:1 To establish animal model:choose 30 adult male wistar rat with similar weight,and randomly divided them into 3 groups,named for the blank group, the control group and model group,each group had 10 rats.blank group:without any treatment,the normal feeding,control group:0.9% saline free drinking 6months,model group:0.02% ammonia free drinking and long-term irregular diet.2 After successfully modeling,all rats were sacrificed and gastric tissues were harvested, stained with hematoxylin and eosin. The expression of Wnt1,β-catenin and Lgr5 at m RNA and protein levels was analyzed by reverse transcriptional polymerase chain reation, westernblotting and immunohistochemisgry respectively.Results:1 HE staining:In blank group and control group,the glandular stomach tissue is normal,there is no abnormal change,but in the model group,there are a lot of inflammatory cell infiltration in gastric antral mucosa muscrlaris propria,muscularis mucosa and submucosa.2 The results of western blot: compared to blank group and control group, Wnt1 and Lgr5 protein levels showed high expression in model group(P<0.05),No obvious difference between normal group and control group was observed(P>0.05).3 The results of immunohistochemistry: Wnt1 and Lgr5 was mainly expressed in the cell membrane and cytoplasm of the cells, Wnt1 and Lgr5 protein levels showed low expression in the blank group and control group, No obvious difference between normal group and control group was observed(P>0.05), compared to blank group and control group, Wnt1 and Lgr5 protein levels showed high expression in model group(P<0.05),β-catenin protein levels showed high expression in the blank group and control group,no significant difference between the two group, compared to the two group, β-catenin protein levels showed high expression in model group(P<0.05).4 The results of RT-PCR: compared to blank group and control group,Lgr5 m RNA levels showed high expression in model group(P<0.05),No obvious difference between normal group and control group was observed(P>0.05).Conclusion:1 Wnt/β-catenin signaling pathway abnormal expression or activated is closely related with the occurrence and development of tumor, In the model group,the expression of Wnt1,β-catenin and Lgr5 was higher than blank group and control group,suggesting that Under inflammatory stimulation,the Wnt pathway may have activated.2 Lgr5 is a kind of target genes of the Wnt pathway, and is closely related to the occurrence and development of tumor, In our study, compared to blank group and control group, Lgr5 protein and m RNA levels showed high expression in model group,so we speculated that Persistent inflammation leads to increased expression of lgr5. Lgr5 may be a tumor promoting factor for inflammation.
