Objective: In order to further discuss the cells significance in ITP pathogenesis, the CD4+CD25+Treg and CD4+ CD70+T cells in idiopathic thrombocytopenic purpura patients as well as the lecel of IL-27 in serum are measured.Methods: 30 cases in our hospital outpatient and inpatient hematology department as ITP patients’ observation group According to the therapeutic effect, the patients are divided into marked effect group and ineffective group after treatment. And 30 healthy human whole blood cases which were validated normally in hospital medical center is as healthy control. Each group is divided into prior treatment and post treatment groups. The post treatment group is divided into marked effect group and ineffective group. The expression percentage of CD4+CD25+Treg cell and CD4+CD70+T cells in prior and post treatment ITP patients as well as in peripheral blood in healthy group are detected by flow cytometry. Using human interleukin-27 ELTSA kit respectively detected the levels of IL-27 in serum of ITP patients before and after treatment and healthy control group. Analysis and compare the percentage of peripheral blood CD4+CD25+Treg cells, the difference of CD4+CD70+T cells positive rates and changes of IL-27 level in ITP patients group and the healthy control group. Further comparatively research the percentage of CD4+CD25+Treg cells, percentage differences of the CD4+CD70+T cells and situation changes of serum IL-27 level in ITP patients in marked effect group and ineffective group before and after treatment. All the test data of the study is analyzed and processed by software SPSS17.0. Normality test adopts the single sample K-S Test. P >0.05 is obeying normal. The data of the two groups in normal distribution adopt the independent sample T test. Multigroup comparison adopts the variance analysis. P <0.05 in difference test is significant difference. All measurement data use the t-test paired t test; qualitative data use the x2 test. And(X± s) is the representation of all data. The α = 0.05 is the test standard and draw the conclusions.Results:1 the percentage of peripheral blood CD4+CD25 +Treg cells expression of ITP patients in the observation group and the healthy control group have statistics different(P<0.05). Either before treatment or after treatment, the expression percentage of CD4+CD25+T cells are lower than the healthy control group(P<0.05), but higher than prior treatment.Comparison between before and after treatment of ITP patients, the expression percentage of peripheral blood CD4+CD25 +Treg cells in ineffective group is lower than that of marked effect group. Both of them have the statistics difference(P<0.05) and are lower than that of the healthy control group(P <0.05).2 The positive rate of CD4+CD70+T cells in the ITP case group patients is higher than that of healthy group, which have the statistics differences(P <0.05). The expression percentage of Peripheral blood CD4+CD70+T cells in ITP patients of ineffective group after treatment is higher than that of post-treatment marked effect group.Both of them have statistics differences(P <0.05), and higher than that of healthy group(P<0.05). Comparison between before and after the treatment itself, CD4+CD70+T cell percentage after treatment compared to prior treatment have the statistics difference(P<0.05). The after treatment is lower than prior treatment, but are all higher than healthy group(P<0.05).3 Serum IL-27 expression in ITP patients plasma levels is higher than that of healthy group, both of them have the statistical difference(P<0.05). Either before treatment or after treatment, IL-27 expression levels are higher than that of healthy group(P<0.05). Comparison between before and after the treatment itself, IL-27 expression levels after treatment is lower than that of before treatment. Both of them have statistics differences(P<0.05).IL-27 expression levels in plasma of ITP patients after treatment is lower than that of after treatment group in ineffective group. both of them have statistics differences(P<0.05).Conclusion: the percentage of CD4 +CD25+Treg cells in ITP patients is lower than that of normal healthy people. With the effective treatment, the percentage of CD4+CD25+Treg cells are increased, but still lower than that of normal healthy people. The expression percentage of CD4+CD70+cells and serum IL-27 levels are higher than that of normal people. With effective treatment, the expression levels decreased, but still higher than that of normal healthy groups. the expression prompt CD4+CD25+Treg cells, CD4+ CD70+T cells and serum IL-27 in the pathogenesis of ITP has a certain significance.
