Background With the development of our national economy and the improvement of living standard,the incidence of atherosclerosis was increasing year by year.There were a variety of theories of atherosclerosis pathogenesis, such as acquired thrombosis theory,lipid infiltration damage response theory, inflammation theory and immunity theory, however the exact mechanism has not yet clear. Recent studies have detected that immune cells played an important role in pathogenesis of atherosclerosis, especially CD4+CD25+Treg cells.However, it still needs further research to exploring the function and impact of CD4+CD25+Treg cells and the related inflammation factors in the process of atherosclerosis.Objective We observed the expression of CD4+CD25+Treg cells and the changes of inflammatory factors through the establishment of the atherosclerosis model in inbred line C57BL/6J mice, to investigate the roles of CD4+CD25+Treg cells and inflammatory cytokines in the atherosclerosis of the inbred line C57BL/6J mice.Methods Twenty-four male inbred line C57BL/6J mice were assigned to three groups:control group, atherosclerosis (AS) group and atorvastatin group.Control group was given normal diet,the rest groups were given high fat diet. After 4 weeks of high fat diet,Atorvastatin group were continued to keep high fat diet and given drugs.Observing the mice for 16 weeks.Vasculopathy was analyzed by Hematoxylineosin staining. Expression of CD4+CD25+Treg in peripheral blood was analyzed by flow cytometry.Concentration of TGF-? and IL-17 was analyzed by Elisa.Results1. As group's content of TC,TG and LDL was significantly higher than control group, (P?0.01).Atorvastatin group's content of TC,TG and LDL was significantly lower than AS group (P<0.01). Both of them were statistical significance.2.1n the normal control group, the artery wall of external and internal elastic plate full, clear, the core of the endothelial cells dyed evenly.The rest of the groups of the artery wall all had different degrees of pathological changes.The group of AS uneven thickening of the vessel wall, vascular wall with inflammatory cells infiltration, lipid deposition.Compared with the AS group, atorvastatin group of vessel wall thickening, a small amount of inflammatory cell infiltration, significantly reduce pathological changes.3.Through flow cytometry, CD4+CD25+Treg/CD4+T cell of AS group was 6.45%±1.24%,which was significantly lower than 10.25%±1.19% of control group(P?0.01). CD4+CD25+Treg/CD4+T cell of Atorvastatin group was 9.75%±1.17%, which was significantly higher than 6.45%±1.24% of AS group(P<0.01). Atorvastatin group and control group had no significant difference.4.As group's Concentration of TGF-? was 8.55±3.26 ng·ml-1,which was significantly lower than 18.32±3.83 ng·ml-1 of control group (P<0.01).Atorvastatin group's Concentration of TGF-? was 16.93±3.67 ng·ml-1,which was significantly higher than that 8.55±3.26 ng·ml-1 of AS group (P<0.01). Atorvastatin group and control group had no significant difference. As group's Concentration of IL-17 was 25.29±5.82 ng·ml-1,which was significantly higher than 15.36±4.67 ng·ml-1 (P?0.01) of control group (P<0.01).Atorvastatin group's Concentration of IL-17 was 18.39±4.98 ng·ml-1,which was significantly lower than that of AS group (P?0.01)Conclusion CD4+CD25+Treg cells may play an important role to suppress the processing of atherosclerosis,which can through secreting TGF-? and inhibiting the secretion of IL-17. The expression of CD4+CD25+Treg cells and the level of inflammatory factors may have much value to predicting the processing of atherosclerosis.
