| Objective: Through the determination of chronic non bacterial prostatitis rat model,and the concentration of Zn of neutrophil CD64 expression in chronic bacterial prostatitis rat model and their false model group, normal prostate tissue, to explore the application value of neutrophil CD64 expression quantity and concentration of Zn detection in the diagnosis of chronic prostatitisMethods:1.Select equal weight(about 200g), 105 healthy SD rats were randomly divided into five groups, For chronic non bacterial prostatitis rat model group(group A), chronic non bacterial prostatitis rat model to group(group B), chronic bacterial prostatitis rat model group(group C), chronic bacterial prostatitis rat model group(D group), normal group(group E), 20 rats in each group, the remaining 5 will be used to extract the immune protein.2. The establishment of the animal model: Establish the rat models of chronic bacterial prostatitis by immunization. Establish the rat models of chronic nonbacterial prostatitis using urethral kinds of bacteria.3.Using a spectrophotometer measure Zn(zinc) concentration in the prostate fluid of 5 rat models. Expression of neutrophil CD64 in the prostate fluid of the 5 groups of rats were determined by flow cytometry.Comparative and analysis the change in the two indicators of Zn concentrations and neutrophil CD64 in A, B, C, D, E rat the prostate fluid.Results: Zn concentrations decreased significantly in the CBP and CNBP model group, With the CNBP module, CBP module, fake fake normal control group there was significant difference(P<0.05). The concentration of Zn in CNBP model group and CBP model group have no significant difference(P>0.05). there is no significant difference between the control group,CNBP fake module and CBP fake module(P>0.05). The expression of neutrophils CD64 was significantly increased in the CBP model group, there was significant difference between the control group,CNBP modules, CNBP fake modules and CBP fake modules,(P <0.05). there is no difference between the control group, CNBP modules, CNBP fake modules, CBP fakemodules in the expression of neutrophil CD64(P> 0.05).Conclusion:1.In this study, immunization, urinary tract bacteria species were successfully constructed two different CP rat model.From the point of view of animal experiments show that autoimmune and bacterial infections may be predisposing factors of CP.2.Neutrophil CD64 can be used to determine whether there is a bacterial infection in the prostate.3.Determination of zinc content can be used to assist in the diagnosis of CP, but it can not distinguish between specific isoforms CP.4.Combined with determination of neutrophil CD64 and zinc content type is effective for the diagnosis of CP. Its conducive to the rational application of antibiotics in the next step. |
