AimGastric cancer as a high incidence of tumor in our country, its incidencespresented a trend of younger age in recent years. But the early diagnosis, treatment and prognosis is still not ideal, and therefore for the early diagnosis and treatment of gastric cancer to find effective biomarkers has become the focus of research at present.RIN1 as an effect factor Ras protein, which can inhibit the activity of Ras.Ras protein plays an important rolein the regulation of cell growth and signal transduction pathway. RIN1 as an effect factor Ras protein, which Participate in a variety of malignant tumors, plays an important role in the development of tumors.SIAH2 as a conserved ubiquitin ligases,which activated by Polyubiquitination of proteins, therefor play a regulating role.The SIAH2 protein can be through a variety of signal transduction pathways involved in tumorigenesis, development, respectively is SPRY2/Ras/ERK and PHD3/HIF-1α. Signal pathway.SIAH2 as a regulatory protein in Ras signal transduction pathway,which participate in a variety of tumor occurrence, development. Schmidt research shows that, the inhibition of SIAH2 protein expression, ras pathway is blocked, tumor growth restricted.Recent research shows that, the up-regulated expression of RIN1 in non small cell lung cancer and gastric cancer are closely related to its clinical pathological characteristics.SIAH2 research is mainly concentrated on in vitro,the reports of RIN1, SIAH2 combined with domesticin gastric cancer are poorly understood.This research through the two combined detection in gastric cancer and normal tissue expression,and to discuss the two role in the carcinogenesis and development of gastric cancer, and provide a theoretical basis for the clinical diagnosis,treatment,and prognosis.Materisls and methods120 archived paraffin specimens were selected from Zhengzhou University First Affiliated Hospita in 2011 June to 2013 December operation resection or endoscopic resection of paraffin blocks,all specimens were pathologically diagnosed.Among the 80 cases of gastric cancer,47 cases of male,33 cases of female.The average age of 54 years, the maximum age of 76 years old, the youngest 34 years old.Histopathological grade:27 cases of high differentiation adenocarcinoma, 53 cases of low differentiated adenocarcinoma.Clinical stages: I / II in 38 cases, 42 cases of stage III / IV. Lymph node metastasis in 33 cases, 47 cases without lymph node metastasis.54 cases of serosal invasion, 26 cases without serosal infiltration.40 cases of normal gastric tissue.All gastric cancer patients before surgery had not received radiation, chemical and immune. All specimens were fixed in 10% formalin, after dehydration, embedded in paraffin, cooling, the specific steps in strict accordance with S-P kit, instead of an anti as blank group with PBS, using known positive sections were used as positive control.The data were analyzed using the SPSS17.0 software, the differences between groups using the Chi-squared test, correlation studies by using Spearman rank correlation analysis, with alpha = 0.05 as the level of test.Results(1) The positive expression of RIN1 protein was located in cytoplasm and nucleus, brown yellow high expression, the expression of SIAH2 protein in the cell nucleus brown yellow particles.(2) In gastric carcinoma,positive rates of RIN1 and SIAH2 proteins were and 61.2% and 60.0%,in normal gastric tissue,the positive rates were 17.5% and 22.5%,both of the results had significant differences(P <0.05).(3) In gastric carcinoma,RIN1 and SIAH2 expression related with the tumor size,clinical stage, histopathological grade, lymph node metastasis and depth of invasion(all P < 0.05),but not related to patients’ age and gender(P>0.05),but the positive expression of RIN1 was related with tumor size(P <0.05)(4) There was a positive correlation between the expressions of RIN1 and SIAH2 in gastric carcinoma(r = 0.607, P <0.01).Conclusion(1) The positive expression rate difference of RIN1 protein in gastric carcinoma and normal tissue has statistics significance.In gastric carcinoma,RIN1 expression related with the tumor size,clinical stage, histopathological grade, lymph node metastasis and depth of invasion. It indicated that RIN1 may play an important in tumorigenesis, development, and associated with tumor invasion, metastasis, which is expected to become a new index judging prognosis of gastric cancer, early diagnosis.(2) The positive expression rate of SIAH2 in gastric cancer was higher than that of normal tissue, the difference has statistics significance.SIAH2 expression related with the clinical stage,histopathological grade, lymph node metastasis and depth of invasion. Therefore, SIAH2 may act as oncogenes promote tumor development and metastasis,which also is expected to become a new index judging prognosis of gastric cancer, early diagnosis.(3) There was a positive correlation between the expressions of RIN1 and SIAH2 in gastric carcinoma.It indicated that RIN1 and SIAH2 has a synergistic effect in tumor occurrence,development, invasion and metastasis, may the existence of common Ras signal transduction pathway.
