ObjectiveGuided by TCM theory, combined with modern medical experimental method,accumulation of damp heat type arthralgia copy of combination of disease and syndrome(acute gouty arthritis) rat model, through the application of traditional Chinese medicine prescription Tongfengkang treatment of humid heat type arthralgia syndrome of acute gouty arthritis in rats were observed after the treatment, rats ankle swelling degree and laboratory check the index(IL-1 beta, TLR4 m RNA,My D88 m RNA, TRAF6 m RNA, NF-kappa B nuclear protein p65) changes, rheumatism analysis Tongfengkang on wet hot type(acute gouty arthritis) in rats, study the possible mechanism of Tongfeng Kang in the treatment towards acute gouty arthritis.Methods72 SD rats were randomly divided into six groups, normal group, model group respectively, Tongfengkang high, medium and low dose group, western medicine group, each group 12 only. Tongfengkang each dose group respectively by Tongfengkang configuration solution with different concentration of gastric perfusion as control group, western medicine group with indometacin gastric perfusion, normal group and model group with physiological saline, every day one time, the course of treaement is 5 days. Medication for third days, in addition to the normal group, all rats were injected by sodium urate solution right ankle joint cavity modeling approach, the establishment of humid heat type arthralgia syndrome(acute gouty arthritis) model in rats. Normal group injected the same volume of saline. Observing rat right ankle joint swelling volume measurement. Enzyme linked immunosorbent assay(ELISA) method was used to detect the content of IL-1 in rat serum beta expression,immunohistochemistry NF- kappa Bp65 in synovial tissue of rats was detected by real-time fluorescent quantitative detection method, the expression of TLR4 m RNA,My D88 m RNA, TRAF6 m RNA, pathological changes were observed in synovial tissuesof rat HE staining method.Results1. To copy the success of heat accumulation type arthralgia syndrome(acute gouty arthritis) model in rats. Compared with the normal group, model group after the model of 4h, 12 h, 24 h, 48 h, 72 h joint swelling was increased(P<0.01). Compared with the model group, Tongfengkang high dose and middle dose group, western medicine group after the model of 4h, 12 h, 24 h, 48 h, 72 h hours of joint swelling was significantly lower(P<0.01); compared with the model group, low dose group after the model of 4h,12 h, 24 h, 48 h, 72 h hours of joint swelling had no difference(P>0.05). Tongfengkang,low dose group and Western medicine group there was a significant difference(P<0.01). No difference to compare Tongfengkang high dose group and Western medicine group(P>0.05), suggesting that Tongfengkang high dose group and Western medicine group equivalent efficacy.2.Normal serum visible little IL-1 beta, compared with the normal group, the serum levels of IL-1 beta in the model group increased obviously, significant differences(p<0.01). Compared with the model group, Tongfengkang high dose and middle dose group and Western medicine group can cause serum IL-1 of rats with acute gouty arthritis( P<0.01), compared with no difference Tongfengkang low dose group with the model group(P>0.05). Tongfengkang low dose group and Western medicine group there was a significant difference(P<0.01). No difference in Tongfengkang high dose group and Western medicine group(P>0.05), suggesting that Tongfengkang high dose g r o u p a n d We s t e r n m e d i c i n e g r o u p h a v e e q u i v a l e n t e f f i c a c y.3.Synovial tissue of normal group had a small amount of NF-κBp65, compared with the normal group, significantly increase NF- kappa Bp65 expression in synovial tissue of model group rats, significant differences(p<0.01). Compared with the model group,NF- significantly reduced kappa Bp65 expression Tongfengkang high, middle dose group, western medicine group rats synovial membrane, significant differences( P<0.01), no difference in the low dose group and the model of Tongfengkang group(P>0.05). Gout, Kang in the low dose group and Western medicine group there was a significant difference(P<0.01). No difference in Tongfengkang high dose group and Western medicine group(P>0.05), suggesting that Tongfengkang high dose group and Western medicine group have equivalent efficacy.4. Little TLR4 m RNA expression was found in normal group joint synovial tissue.Compared with the normal group, model group rats synovial tissue in TLR4 m RNA relative expression quantity increased obviously, significant differences(P<0.01).Compared with the model group, high dose group, Tongfengkang, the western medicine group rats synovial membrane of TLR4 m RNA relative expression quantity obvious reduction, significant differences(P<0.01), no difference in the low dose group and the model of Tongfengkang group(P>0.05). Tongfengkang in the low dose group and Western medicine group there was a significant difference(P<0.01). No difference in Tongfengkang high dose group and Western medicine group(P>0.05),suggesting that Tongfengkang high dose group and Western medicine group equivalent efficacy.5. A small amount of TRAF6 m RNA expression in normal group joint synovial tissue,compared with the normal group, model group rats synovial tissue in TRAF6 m RNA relative expression quantity increased obviously, significant differences(P<0.01).Compared with the model group, high dose group, Tongfengkang, the western medicine group rats synovial membrane of TRAF6 m RNA relative expression quantity obvious reduction, significant differences( P<0.01), no difference in the low dose group and the model of Tongfengkang group(P>0.05). Tongfengkang in the low dose group and Western medicine group there was a significant difference(P<0.01). No difference in Tongfengkang high dose group and Western medicine group(P>0.05),suggesting that Tongfengkang high dose group and Western medicine group equivalent efficacy.6. A small amount of My D88 m RNA expression in normal group joint synovial tissue, compared with the normal group, model group rats synovial tissue in My D88 m RNA relative expression quantity increased obviously, significant differences(P<0.01).Compared with the model group, high dose group, Feng Kang in, the western medicine group rats synovial membrane of My D88 m RNA relative expression quantity obvious reduction, significant differences(P<0.01), no difference in the low dose group and the model of Tongfengkang group(P>0.05). Gout, Kang in the low dose group and Western medicine group there was a significant difference(P<0.01). No difference in Tongfengkang high dose group and Western medicine group(P>0.05), suggesting that Tongfengkang high dose group and Western medicine group have equivalent efficacy.7. HE staining of model group rats disease obvious synovitis, synovial cell hyperplasia,accompanied by inflammatory cell infiltration, vascular proliferation and fibroblasts,suggesting the model is successful, the Western Medicine(colchicine) and Tong feng Kang after the treatment, the pathological changes alleviated.Conclusion1.Establishment of a model of acute gouty arthritis in rats using the modified coderre[1]method, basically reflects the humid heat type arthralgia symptoms, syndrome of damp hea t ac c u m u l a t i o n ty p e ha s th e ch a r a ct er i s t i c s of th e mec h an i s m.2. Gout Concord improve sodium urate crystal joint swelling induced acute gouty arthritis model rats, improve the pathological changes of synovial tissue.3. Gout Concord down content in serum of rats with acute gouty arthritis model of IL-1β, down regulate the expression of synovial tissue of acute gouty arthritis in the rat model of TLR4 m RNA, My D88 m RNA, TRAF6 m RNA, NF- κBp65 of the TLR4/NF-κB pathway intervention, may be the Tongfengkang treatment of acute gouty arthritis is one of the mechanism.
