Objective:To take spontaneously hypertensive rats for the study, observing the acupuncture effecs on the Yuan-Sourse acupoint Of kindy Taixi point and t he acupuncture point without going through the regulate channals on blood pressure of SHR. Analyzing the expression of CRP and IL-6 in hippocampus and the medulla oblongata by immunohistochemistry in spontaneously hyper tensive rats’s inflammatory cytokines. To explore the action of acupunctur e Taixi Point in essential hypertension.Methods:Use Thirty 11-week-old male spontaneously (SHR) model of hypertensive rats as experimental subjects. After adaptive feeding one week, divide t he 30 SHR into three groups randomly, named acupuncture Taixi Group, Non-acupuncture point group, model control group,n=10. Acupuncture and bloo d pressure measurement must be conpleted every day, keep treating for 7 d ays, choosing blood pressure as the main indicator. The next day after th e end of treatment, all the rats were decapitated. Then extract hippocampu s, medulla oblongata in time, immunohistochemical analysis methods were u sed to analyze the optical density and positive area of C-reactive protei n(CRP), interleukin-6 in hippocampus and medulla oblongata as experiment s chamber indicators. Compairing the blood pression of each group rats, be fore and after acupuncture treatment, then clarify acupoint specificity. In addition, by comparising the expression levels of medullary C-reactive protein(CRP), interleukin-6(IL-6) in hippocampus and medulla oblongata o f rats, to explore the possible mechanisms of hypertension on acupuncture Taixi Point.Results:1.After seven day’s treeting, be compaired with the SHR model group, T he blood pressure of acupuncture Taixi group lowered obviously, (P<0.05), there was significant difference (P<0.05); The blood pressure of non- a cupuncture points did not change acommonly, there was no significant diffe rence (P>0.05)2. Immunohistochemical results:(1) The expression of IL-6in the medulla oblongata:Compared with the model group, acupuncture Taixi group, the optical density and positive s taining area of IL-6 in medulla oblongata slices was greatly reduced, sta tistical significant difference (P<0.01); Compared with acupuncture Taix i group, acupuncture non-acupoint group’ soptical density and positive are a increased, the difference was statistically significant (P<0.05).(2) The expression of IL-6 in the hippocampus:Compared with the mod el group, acupuncture Taixi group, the optical density and positive stain ing area of IL-6 in hippocampus slices was greatly reduced, statistical s ignificant difference (P<0.01); Compared with acupuncture Taixi group, ac upuncture non-acupoint group’ soptical density and positive area increased, the difference was statistically significant (P<0.05).(3) The expression of CRP in the medulla oblongata:Compared with th e model group, acupuncture Taixi group, the optical density and positive staining area of CRP in medulla oblongata slices was greatly reduced, sta tistical significant difference (P<0.01); Compared with acupuncture Taixi group, acupuncture non-acupoint group increased, the difference was stat istically significant (P<0.05).(4) The expression of CRP in the hippocampus:Compared with the mode 1 group, acupuncture Taixi group and acupuncture non-acupoint group, the optical density and positive staining area of CRP in medulla oblongata sl ices didn’t appear greatly reduced, statistical difference was not signif icant (P>0.05); Compared with acupuncture Taixi group, acupuncture non-ac upoint group mearly increased, the statistical difference was not signifi cant (P>0.05).Conclusion:1. acupuncture Taixi point can significantly reduce blood pressure in spontaneously hypertensive rats,and has effective treatment.2. Immunohistochemical staining methods analyze optical density rate and the rate of positive area of IL-6 and CRP in hippocampus and medulla oblongata for each group SHR. The results suggest, acupuncture Taixi point, expression levels of IL-6 in the medulla oblongata and hippocampus has si gnificantly reduced, the expression of CRP in the medulla oblongata levels were significantly lower, but expression levels of CRP in the hippocampus altered unobviously. It describe that acupuncture Taixi point has a certa in effect on the expression of central inflammatory factors in SHR.
