Object Probe into the molecular evolution of IL-33 gene family in different species, further illustrate its impact in the reaction of allergic rhinitis. Investigate the expression changes of IL-33 and relevant genes in the process of treatment. Through correlation analysis of expression patterns, the mechanism of Nasal Oil in the process of treatment is clarified,besides, the clinical research and drug development foundation of Nasal Oil is also laid.Methods1 、 Bioinformatics analysis : Identification of IL-33 genes and construction phylogenetic trees of human IL-33 proteins and classification, property analysis of Human IL-33 protein amino acid sequences of the gene encoding sequeces, replication of the human IL-33 protein genes, chromosome physical location of Human IL-33 protein genes, exon and intron analysis of Human IL-33 protein structure,analysis of Human IL-33 protein genes conserved motifs, completely conserved sequence motif analysis of all IL-33 proteins used Weblogo software, calculation of Ka and Ks---inferred IL-33 paralogous genes in humans from phylogenetic trees2、Analysis of gene expression patterns. April- May 2014,60 cases of patients with allergic rhinitis were selected from First Affiliated Hospital of Anhui University of Chinese Medicine, otorhinolaryngology. Randomly separated into two groups, treatment group and control group,before treatment, appropriate amount of nasal mucosa was applied to all patients, RNA was extracted. Therapeutic group was treated with nasal nasal drip, 3 drop in each lateral nasal(10 ul), three times every day; budesonide nasal spray was used in control group, 1 in each lateral nasal spray at a time(0.280mg) three times a day, four weeks for 1 course of treatment for both of the two sets, after one couse of treatment, take the nasal mucosa of all patients, extract RNA. RT-PCR nd fluorescence Quantitative PCR of human IL-33 gene, The before and after treatment IL-33 of budesonide group and Xinyi nasal drops were compared, Effects of compound Xinyi nasal drops on clinical curative effect of patients with allergic rhinitis, andwhether or not significant difference of the IL-33 were observed before and after treatment in patients. Results show the two groups in the course of treatment is safe and reliable; Test group and the control groupis are effectly.Conclusions(1) Based on the Molecular Evolution of IL-33 gene family in different species, a high similarity was found in Human and mouse chromosome.Research results on the IL-33 in mice may also apply to humans.(2)Though IL-33 and its related studies of gene expression changes during the treatment, when compared to an immune response improvement, their total amount of immune response occurrence is found to be reduced.Thus, we can infer that part of the IL-33 were consumed in the immune response.(3)PCR analyze the change in amount of IL-33 in measure and Control Group before and after treatment of AR nasal drops. Budesonide Group, higher than the nasal drops group,higher than saline control group. The results shows that Xinyi nasal drops have certain clinical curative effect on allergic rhinitis patients, IL-33 expression was significantly decreased. In the IL-33 involved pathogenesis of allergic rhinitis, measurement of IL-33 were able to objectively evaluate the effect of allergic rhinitis, providing a new way of thinking Diagnosis and treatment of allergic rhinitis...
