ObjectivesThrough analysis of gene polymorphism, understand general clinical features, reproductive hormones and related biochemical indicators of the south area of the Fujian PCOS, investigate the correlation between polymorphism of LEPR gene G1n223Arg and polycystic ovary syndrome (PCOS).MethodsThe research methods was case-control study. Using a questionnaire survey to collect epidemiological data of PCOS women and collect the venous blood.100 patients with PCOS women(Rotterdam diagnostic criteria (2003)) as the experimental group, 154 cases of chronic salpingitis women as control group. The leptin receptor (LEPR) gene polymorphism was examined by polymerase chain reaction (PCR) and direct sequencing.Results1. In PCOS group, compressive, abdomen circumference, hairy, acne, LH, LH/FSH, T, fasting insulin, triglycerides, high-density lipoprotein cholesterol, HOMA-IR increased significantly, and the difference was statistically significant.2. In PCOS group, the homozygous mutant GG genotype was 87 cases, heterozygote AG genotype was 13 cases; In control group, the wild homozygous genotype AA wasl cases (0.7%), homozygous mutant GG was 120 cases (77.9%), heterozygous AG genotype was 33 patients (21.4%). P= 0.069 tested with Chi-square test, So it was found no statistically difference between PCOS and control group of AG and GG genotype.3. In PCOS group, the HOMA-IR of genotype (AA + AG) was higher than that of genotype GG (P< 0.05).ConclusionsLEPR gene G1n223Arg polymorphism has no direct relevance with susceptibility of PCOS. Consequently, we predicted that LEPR gene G1n223Arg mutation was the risk factors for insulin resistance, and indirectly leads to the occurrence of PCOS.
