The Expression Of Galanin In Hippocampus And Colon Under Chronic Unpredictable Stress And The Effect Of Kaiyuanshen Decoction

Objective:1.To observe the effect of Kaiyu Anshen Decoction by behavioristics, morphological change and expression of galanin in hippocampus and colon in depression model rats, to explore the pathogenesis of depression and its concomitant symptom of digestive tract;2.By observing the clinical effective rate of Kaiyu Anshen Decoction in the treatment of depression to provide better scientific basis for the popularization and application of Kaiyu Anshen Decoction.Methods:1.Experimental study: 60 adult female SD rats were selected and divided into two parts,control group and big model group. Using the chronic unpredictable mild stress combining with feeding lonely to establish the model of depression.Then choose those meeting a criterion of concomitant symptom of digestive tract to enter the experiment.Divide them into three groups randomly: model group, fluoxetine group and Kaiyu Anshen Decoction group. Administrating drugs while establishing model.Weight growth、sucrose consumption, horizontal and vertical score of open field test,change of first defecating time were observation target.Cellular morphology of hippocampus and colon were observed under the optical microscope and content of galanin in hippocampus and colon were measured with immunohistochemical method. 2.Clinical study: 62 patients with depression were selected.They were randomly divided into two groups,32 cases of Chinese medicine group, 30 cases of western medicine group.The clinical effective rate was judged by HAMD, SDS, SAS and the Chinese medicine symptom scale.Results:1.Experimental research:(1) Horizontal and vertical score of Kaiyu Anshen Decoction group were significantly higher than those in model group;sucrose consumption decreased significantly;weight growth slowed down significantly.Compared with fluoxetine group,there was no significant differrence.(2)Compared with fluoxetine group, chang of first defecating time of Kaiyu Anshen Decoction group had significant difference.(3)In hippocampus, positive cell numbers and average gray value of galanin of model group was significantly higher than the control group,Compared with the model group,Kaiyu Anshen Decoction and fluoxetine group decreased significantly.(4)In colon,positive cell numbers and average gray value of galanin of model group was significantly higher than the control group;Compared with model group,Kaiyu Anshen Decoction and fluoxetine group decreased significantly. This indicates that galanin play different role under different conditions. 2.Clinical research:(1)The total curative effect of traditional Chinese medicine group was 86.67%,western medicine group was 89.29%, there were no significant difference. Comparing traditional Chinese medicine group with western medicine group, the reduction rate of HAMD,SDS,SAS had no significant difference.(2)For the Chinese medicine symptom scale,the overall score of traditional Chinese medicine group and western medicine group had no significant difference,but in the improvement of somatic anxiety symptoms and gastrointestinal symptoms, traditional Chinese medicine has obvious advantages than western medicine.This indicates that Kaiyu Anshen Decoction is better at curing depression of Liver qi stagnation and heart fire flaming up. Kaiyu Anshen Decoction also has advantage at gastrointestinal symptoms.Conclusion:1. Galanin may participate the occurrence and development of depression and its concomitant symptom of digestive tract.2. Kaiyu Anshen Decoction can adjust the therapeutic effect on rat model of depression associated with gastrointestinal symptoms.3.For general symptoms of depression,the effect of Kaiyu Anshen Decoction and fluoxetine is almost identical, while for gastrointestinal symptoms, Kaiyu Anshen Decoction is better.
