Backgrounds and objective:Sepsis is defined as the presence of systemic inflammation response syndrome(SIRS) that caused by microbial infection. Its pathogenesis is not completely clear, and there are a large number of studies have showed that it is associated with genetic factors.IL-10 is a inflammatory cytokines and it have various functions. It plays anti-inflammatory efficacy by reducing the mononu-clear macrophage activity and antagonism of inflammatory mediators. Previous study showed that the IL-10 gene polymorphisms involved in the whole process of sepsis’s pathogenetic process, and it played a protective role in the occurrence and development of sepsis. At persent the research about IL-10 gene and the relationship between its polymorphism and sepsis is less. According to analysis the distribution of IL-10 alleles and genotypes in the Guangxi population, we aim to investigate the relationship between IL-10 SNP and sepsis susceptivity.Method:We picked up 152 patients with sepsis in ICU of Guangxi as the case group,199 healthy people over the same period are recruited as the control group. In order to evaluate whether the three SNPs in IL-10 gene were associated with sepsis susceptibility, the polymerase chain reaction-single base extension (PCR-SBE) and DNA sequencing were used to research the SNPs of IL0-10(rs1800871、rs3021094、rs1800872) in sepsis in the Guangxi population.Results:There were no significant differences in gender and age between the case group and control group. Compare to the control group, rs3021094 genotype distribution was significantly associated with the risk of sepsis (P=0.028). In the case group, the frequencies of T, G alleles at IL-10 gene promoter rs3021094 position were 41.4% and 58.6%; in control group the frequency distribution were 51.5% and 48.5%, The frequency of risk allele of rs3021094-G in the case group was notably higher than that of controls (OR: 1.501; 95%CI:1.110-2.028; P=0.008),it suggest that polymorphism in rs3021094 was significantly associated with the risk of sepsis. The frequencies of the other two SNPs in IL-10(rs1800871、rs1800872) did not have significant statistical difference in the two groups. The serum level of IL-10 that expressed in case group is obviously higher than the control group(P=0.000).The serum level of IL-10 is Associated with the genotype of rs3021094, GG and GT genotypes of rs3021094 may increase the serum level of IL-10 (P<0.05).Conclusion:1. In the Guangxi population, the rs3021094 polymorphism is Associated with sepsis susceptibility. The G allele may be a risk factor in the pathogenesis of sepsis.2. In the Guangxi population, the serum level of IL-10 increased significantly, GG and GT genotypes of rs3021094 may increase the serum level of IL-10.GG and GT genotypes may be the risk factors of sepsis.3. The result suggest that rs1800871 and rs1800872 polymorphisms are not associated with sepsis in the Guangxi population。...
