Objective:To study the frequencies of allele and genotype ofinterleukin-6(IL-6) rs1800796C/G in the promoter region in the populationsof Guangxi and the patients with sepsis, and to explore the relationshipbetween IL-6rs1800796C/G gene polymorphisms and sepsis.Methods: The polymerase chain reaction-single base extension (PCR-SBE)technique and DNA sequencing were used to measure the rs1800796C/Gpolymorphism in the IL-6promoter region in199Guangxi populations and152patients with sepsis. Serum IL-6was detected by flow cytometry.Results: Alleles G、C and genotypes GG、GC、CC were found in IL-6genepromoter region rs1800796site in Guangxi populations. There was nosignificant difference of the frequencies of allele and genotype of rs1800796C/Gpolymorphism between male and female group (P>0.05) in Guangxipopulations, and in the distribution of two allele in healthy population andpatients with sepsis (P>0.05). No significant difference of the distribution ofallele and genotype frequencies of rs1800796C/G polymorphism were foundwhen compared with HapMap-HCB and HapMap-JPT SNP data (both P>0.05),but there were significantly difference when compared with HapMap-CEU andHapMap-YRI (both P<0.05). CC genotype was accounted for a higherproportion in the populations of Guangxi, HapMap-HCB and HapMap-JPT,while the HapMap-CEU and HapMap-YRI had a higher frequency of GG type. Serum IL-6increased significantly in patients with sepsis (P<0.05), but nocorrelation was observed between IL-6rs1800796C/G polymorphism and serumIL-6.Conclusion: The genotype distribution and allele frequency distribution ofIL-6rs1800796C/G in the promoter region in the populations of Guangxi haveno correlation with gender, but there were significant difference comparedwith other ethnic groups. No correlation was observed between the IL-6rs1800796C/G gene polymorphisms and susceptibility of sepsis and serum IL-6.
