Backgrounnd and ObjectiveEndometrial polyp (EMP),is a common disease which resulting in abnormal vaginal bleeding,It can occur at any age after the menarche,the incidence of it is about 25%,the peak age is 50years old.Along with the increased abilityl of supplementary examination、the spread of transvaginal ultrasonography and hysteroscopy, the detection rate of endometrial polyp is increased,but,until now,the pathogenesis of endometrial polyp is uncertained,previous studies thround that endometrial polyp belongs to chronic inflammation, some scholars believe that the formation is related to the endocrine state of human body, recent studies propose that the formation of endometrial polyp is related to apoptosis regulation disorder. Most of endometrial polyp are benign lesion, but have a certain risk of malignant transformation, although some scholars believe that the risk of endometrial polyp malignant is low, along with modern living standards will continue to improve, women’s awareness of health care strengthen, more and more peri-menopausal women choose hormone replacement therapy to prevent and cure the peri-menopausal syndrome, thus, the incidence of endometrial polyps malignant will increase,the relevant factors of malignant change was extensive researched by scholars all over the world in these years, is it a kind of precancerous lesion just like rectal polyps is unknow.PTEN(phophatase and tensin homolog deleted on chromosome 10) is a tumor suppressor gene with dual specificity phosphatase activity,it can regulate the proliferation and apoptosis、adhesion and metastasis of cell by protein phosphatases and lipid phosphatase pathway.There are loss of PTEN expression in a variety of malignant tumors including uterine endometrial carcinoma,the mutation rate of PTEN in endometrial carcinoma is about 32%-83%,which is higher than either the mutation rate of c-erbB-2、p53 or K-ras in endometrial carcinoma.Gene mutation of PTEN in endometrial carcinoma may play an important role in the carcinogenesis mechanism. The proto oncogene c-erbB-2 is one members of epidermal growth factor family, its overexpression play an important role in the occurrence of tumor, inhibition of apoptosis, cell migration and angiogenesis. This study was detecte the expression of PTEN gene and c-erbB-2 gene protein in endometrial polyps by immunohistochemistry,to learned the meaning of PTEN gene protein and c-erbB-2 gene protein in it,and analysy the development trend of endometrial polyps in patients with his clinical cases features.Materials and methods1.The object of study:choose 178 endometrial tissue of the patient who had a treatment in the First Affiliated Hospital of Guangxi Medical University from january of 2006 to august of 2013,all tissues were confirmed by pathological examination, there are 86 endometrial polyps,31 atypical endometrial hyperplasia,31 endometrial carcinona,30 normal endometrial tissue.2.Method:detecte the expression of PTEN gene and c-erbB-2 gene protein in endometrial tissue with immunohistochemical (streptavidin-perosidase) way.Use spss 16.0 software package for data processing.Results1. The negative expression rates of PTEN in normal endometrial tissue group were significantly lower than these in endometrial polyps、atypical endometrial hyperplasia and endometrialcarcinona groups(p<0.01).2. The positive expression rates of c-erbB-2 in normal endometrial tissue group were significantly lower than these in endometrial polyps、atypical endometrial hyperplasia and endometrialcarcinona groups(p<0.01).3. The negative expression rates of PTEN in endometrial polyps、atypical endometrial hyperplasia and endometrialcarcinona groups were showing a increasing trend, there were no significantdifference among the endometrial polyps, atypical endometrial hyperplasia and endometrialcarcinona groups(p>0.05).4. The positive expression rates of c-erbB-2 in were no significantdifference among the endometrial polyps, atypical endometrial hyperplasia and endometrialcarcinona groups(p>0.05).5. The expression of PTEN in mild and moderate atypical endometrial hyperplasia groups had a significant difference than that in severe atypical endometrial hyperplasia groups.6.Endometrial polyps with vaginal bleeding have a high negative expression rates of PTEN.7.Endometrial polyps with vaginal bleeding have a high positive expression rates of c-erbB-2. Conclusion1.Endometrial polyp have malignant risk, the endometrial polyp patient with vaginal bleeding is the high risk group of endometrial polyp malignant;2.PTEN is involved in the development of endometrial polyps;3.c-erbB-2 is involved in the development of endometrial polyps.
