Objective:To investigate the expression of PTEN and c-erbB-2 in endometrial carcinoma tissue and to explore its correlation with clinical behavior and pathological characters,as to investigate the relationship between both PTEN and syndecan-1 and the development,the clinical significance as well as the mutual relationsMethods: A total of 52 specimens of endoumetrial carcinoma were evaluated by immunohistochemical technique to detect PTEN and c-erbB-2.Twenty normal endometria and twenty eight atypical hyperplasia endometria were used for control.Result:1. The ratio of PTEM expression deletion in Endometrial cancer is obviously higher than the ratio of endometrial atypical hyperplasia and normal endometrium, with the ratio of PTEM expression deletion is 55.77%,31.03% and 5.56% respectively. Through statistical analysis, it is found that the ratio of PTEM expression deletion has significant statistical difference among the four group(X2 =15.322, p<0.001).2. It is also discovered that the ratio of PTEM expression deletion is related to the encometrial carcinoma pathological grade. The ratio of PTEM expression deletion in poorly differentiated endometrial cancer is greater than that in well-differentiated endometrial cancer,with the ratio of PTEM expression deletion (G1,G2,G3) of 35.00%,63.64%,80.00% respectively. But the menstrual status of patients,clinical stage,muscular layer infiltration and Lymph node metastasis have no obvious correlation with PTEN protein expression and the lack.3. The ratio of PTEM expression deletion in atypical hyperplasia group is 53.85%,which is higher than that in compoud hyperplasia group and Simple hyperplasia group .In endometrial atypical hyperplasia group , there is a positive correlation between expression of PTEN loss and the The degree of hyperplasia. And the expression of PTEN loss of stage I,II,III are 12.5%, 12.5% and 53.85%(p <0.05).4. The positive rates of c-erbB-2 in endometrial cancer tissue is markedly greater than those in endometrial atypical hyperplasia and normal endometrium(p<0.05).5. The positive rates of c-erbB-2 is correlated to clinical stage,muscles invasion and lymph node metastasis (p﹤0.05) but not related with menstrual status of patients(p﹥0.05).6. The expression of PTEN protein were negatively correlated with c-erbB-2 protein .Conclusion:1. Loss of PTEN protein and C-erbB-2 protein overexpression may play an important role in the genesis and development of endometrial carcinoma.2. It is obvious that The positive rates of PTEN in in endometrial cancer tissue (55.77%)is lower than those in endometrial atypical hyperplasia and normal endometrium. At the same time,there is a decreasing trend of PTEN expression with the increased endometrial atypical hyperplasia grade. Loss of PTEN expression is an early event in tumorigenesis of endometrioid endometrial adenocarcinoma.It can be used as the early diagnosis of endometrial cancer marker.3. The expression of PTEN protein were negatively correlated with pathological grade.our data suggest that low and loss of PTEN expression show higher degree of malignancy in endometrial cancer.4. The positive rates of c-erbB-2 in endometrial cancer tissue is markedly greater than those in endometrial atypical hyperplasia and normal endometrium.The expression of c-erbB-2 protein is positively correlated to clinical stage,muscles invasion and lymph node metastasis.5. Binded detections of PTEN and C-erbB-2 protein have positive effect onearly diagnosis , prognosis assessment of endometrial carcinoma, and provide clinic treatment with indicators.
