| Candida albicans is one kind of conditional pathogenic fungus which exists widely in nature and mouth, respiratory tract, digestive tract and vagina mucosa in human body. Candida albicans will not lead to disease at the normal circumstance. However, when the balance of bacterial flora in the body is destroyed by some factors, candida albicans will turn into pathogenic bacteria which will grow fast at a large scale in part of body, causing local even systemic disease. In recent years, candida incidence of infections are rising gradually as the result of the broad applications of antibacterial drugs, hormone and immune inhibitors; the popularity of interventional surgery and the yearly increase of disease risk such as malignant tumor and AIDS. At present, Azoles are the first choices for clinical treatment and prevention of candida infection. But the large use of azoles not only can cure the candida infection, but also increase the drug-resistance of candida especially candida albicans. Therefore, it’s urgent to find new drugs which are high-efficiency, safe and not easy to produce drug-resistant strains.The traditional Chinese herbal medicine are rich in China. It has been found during long time of research that many Chinese traditional medicine(TCM) have the function of the resistance of candida albicans. And some has significant effect even in low concentration. Therefore, the study on the inhibition of the traditional Chinese medicine(TCM) to candida albicans has an positive significance for the clinical medication of candida albicans.Objective:To investigate the seven kinds of traditional Chinese medicine evodia rutaecarpa, honeysuckle, stellera, rhizoma coptidis, fructus schisandrae, radix angelicae sinensis, tougucao inhibition of oral candida albicans; Identify of 75% ethanol extract of seven drugs under different concentrations inhibitionrate,in order to supply the experimental basis for drug resistance of candida albicans for traditional Chinese medicine research and development to provide experimental basis.Methods:1 Extract seven kinds of Chinese medicine.60g of drying,crushing seven kings of Chinese herbal medicine were put into 1000 m L volumetric flask respectively, soaked with 600 m L 75% ethanol solution for 1.0h, 81 ℃water bath circumfluence extraction for 1.0h, repeat three times of extraction, merging the extracted liquid, vacuum filtration, the filtrate respectively and vacuum distillation to no alcohol taste, concentrate with distilled water to constant volume was 2g/m L crude drug. Cryogenic centrifugal, take on clear liquid, with 0.22 mu m aperture of the bacteria filter filter after extract bacteria in brown bottles, in the refrigerator for 4 ℃in standby.2 Seven kinds of Chinese medicine extract in vitro bacteriostasis experiment of experimental strains are dental clinical strains of candida albicans. The modified experimental times test dilution method wad used to detect Inhibitory effect of seven kinds of Chinese herbal medicine 75% ethanol extract on oral Candida albicans. 5 small sterile tube as a group, marked respectively 1, 2, 3, 4, 5; a total of 23 groups. The final concentrations of the drug in each tube are: 1g/ml, 0.5g/ml, 0.25g/ml, g/ml 0.125, 0.0625g/ml. The same preparation method of 23 groups of test tube, five in each group, the experiment will be 23 kinds of experimental bacteria liquid are joining the 23 groups containing liquid medium in test tubes, each tube of 0.1ml bacteria liquid. Marked respectively on the other three tubes as "savarygilliard broth control", "drug concentrate control", "test bacteria growth". The above is an experimental drug model, also set up the rest of the six drugs in experimental models. All the test tubes are placed in 37 ℃constant temperature box, and observed after 24 hours’ cultivation. In charge of meet the requirements of the case, the tablet will each experimental tube cultures inoculated into Sabouraud agar, determine whether the growth of bacteria. 37 constant temperature box, observe the results after 24 h cultured in agar plate, completely no bacterial growth as the standard, record the experimental data, the calculation of seven kinds of Chinese herbal medicine 75% ethanol extract in not through the inhibition of 23 strains of Candida albicans clinical strain rate concentration.Results:The concentration of 75% ethanol extract of seven kinds of Chinese medicine were 1.0 g/m L, 0.5 g/m L, 0.25 g/m L, 0.125 g/m L, 0.0625 g/m L oral candida albicans nhibition rate is respectively L:Fructus schisandrae: 91.30%, 17.39%, 8.70%, 4.35%, 4.35%Angelica: 73.91%, 17.39%, 4.35%, 4.35%, 4.35%Rhizoma coptidis: 65.22%, 30.43%, 13.04%, 8.70%, 4.35%Stellera: 43.48%, 13.04%, 8.70%, 4.35%, 4.35%Evodia rutaecarpa: 43.48%, 13.04%, 13.04%, 8.70%, 4.35%Honeysuckle: 20.09%, 8.70%, 8.70%, 4.35%, 4.35%Tougucao: 13.04%, 4.35%, 4.35%, 4.35%, 4.35%Seven kinds of traditional Chinese medicine(TCM) in different concentrations of 75% ethanol extract of oral candida albicans strains 4 clinical beads bacterium inhibition rate was 100%.Evodia rutaecarpa 75% ethanol extracts in various concentrations of 5 strains of oral candida albicans clinical strains of inhibition rate was 100%.Conclusions:1 Bacteriostatic test in vitro showed that, Wu Zhuyu, honeysuckle, Stellera chamaejasme, Coptis chinensis, Fructus schisandrae chinensis, angelica, Speranskia grass of seven kinds of Chinese herbal medicines have different degrees of inhibition of oral Candida albicans, and showed a concentration dependent. Inhibitory effect: fructus schisandrae > angelica >rhizoma coptidis > stellera evodia 、rutaecarpa > honeysuckle > tougucao.2 The fourth strains of Candida albicans are highly sensitive to seven drugs studied are presented on each concentration effect, indicating that they are derived from the sensitive body. |
