Objective To investigate the effect of sodium tanshinone IIA sulfonate on expression of CX3Cchemokine receptor 1 (CX3CR1)、nuclear factor kappa B(NF-κB)and tumor necrosis factor α (TNF-α)mRNA in a rat model of chronic neuropathic pain.Methods Fifty-four male Wistar rats(180~220 g),aged 6-8 weeks, randomly ,rats were divided into 3 groups, each group contains 18 rats:sham operation group (group S):we just expose the right sciatic nerves but do no ligation, neuropathic pain group (group CCI) and group Sulfotanshinone Sodium Injection (SSI):in accordance with Bennett’s method,we place four ligatures around the right sciatic nerve group. In groupSSI,SSI 25 mg/kg was injected intraperitoneally once a day starting from the end of operation until one day the animals were sacrificed,while in groups S and CCI, the rats received the equal volume of normal saline instead of SSI.Mechanical withdrawal threshold (MWT) and thermal withdrawal latency (TWL) were measured on 1 pre-operative and the 3rd,7th,14th d after operation. After the measurement of pain threshold in each group on the 3rd,7th,14th d after operation,six animals were randomly killed and the L4-6 segments of spinal cords were removed.The expression of CX3CR1,NF-κB and TNF-amRNA was assessed by realtime fluores-cence quantitative PCR (FQ-RT-PCR).Results Compared with group S,MWT and TWL in CCI and SSI were significantly lower on each post-operation and the expression of CX3CR1,NF-κB and TNF-α mRNA.After operation at T1~3 in group CCI and SSI, CX3CR1.NF-κB and TNF-a mRNA were markedly increased (P<0.05). Compared with group CCI,TWL and MWT were markedly increased, the expression of CX3CR1,NF-κB and TNF-a mRNA were markedly down-regulated at T1~3 of post-operation in group SSI(P<0.05). Compared with Ti, the expression of CX3CR1mRNA were down-regulated ,but the expression of NF-κB and TNF-amRNA were increased at T2~3 of post-operation in group CCI and SSI (P<0.05). Compared with T2 the expression of CX3CR1,NF-κBandTNF-amRNA were decreased at T3 of post-operation in group CCI and SSI (P<0.05). Conclusion:SSI can ameliorates chronic neuropathic pain of rats is related to inhibition the mRNA of CX3CR1 and TNF-a induced by the NF-κB activation.
