In this thesis, Traditional Chinese Medicine (TCM) extract tanshinoneII-A has been chosen to study its effect on the expressions of IL-6 gene and IL-10 gene in RAW264.7 cell line according to MTT assay and quantitative RT-PCR. The results obtained from these experiments are as follows.Firstly, the RAW 264.7 cells being treated with lipopolysaccharide (LPS) for 6 hours to induce the inflammation, then be treated with different concentrations of tanshinoneⅡ-A, after being inhibited for 24 hours, 48 hours, MTT assay was used to measure results of RAW 264.7 cells. The results showed that tanshinoneII-A have no effect on RAW264.7 cell line after 24 hours, but it inhibits the cell line obviously after 48 hours. In MTT assay, It had been evaluated that IC50 of tanshinoneⅡ-A which is 48.6μM.Secondly, the total RNA was extracted from the RAW 264.7 cells which being treated with lipopolysaccharide (LPS) for 6 hours and different concentrations of tanshinoneⅡ-A for 48 hours .Than a quantitative RT-PCR was performed to obtain the bands of IL-6,IL-10 andβ-actin genes'fragments by agarose gel analysis. The results showed that tanshinoneⅡ-A can remarkably inhibit the expression of IL-6,while enhance the expression of IL-10 obviously. The expression of IL-6 gene were decreased from the total value to 53.75% and 60% with stimulating by 30μM tanshinoneⅡ-A and 40μM tanshinoneⅡ-A . The expression of IL-10 gene were increased from the total value to 200% and 500% with stimulating by 30μM tanshinoneⅡ-A and 40μM tanshinoneⅡ-A .As discussed above, by inhibiting the expression of IL-6 gene and enhancing the expression of IL-10 gene, tanshinoneⅡ-A has effect of anti-inflammatory. The mechanism is similar to the common anti-inflammatory. This research supplied some new proof and academic basis for developing tanshinoneⅡ-A as a new anti-inflammatory.
