ObjectiveTo investigate the express of CX3CR1 in mouse ischemic stroke animal model of neurons at different time-point and different parts, to provide new ideas and methods for the treatment of ischemic stroke.MethodsThe establishment of animal model with middle cerebral artery occlusion(MCAO), dynamic observation of postoperative symptoms of different groups of mice and Longa score; Using paraformaldehyde perfused brain in 06 hours,12 hours,24 hours,48 hours,72 hours after modeling. Distribution of immunohistochemical observation in different parts of the brain in different groups,expression CX3CR1 of positive neurons; cells were identified by immunofluorescence method; to observe the changes of cerebral infarction area of different groups and calculate the infarct area ratio.Results1. By double staining show, In the animal model of ischemic stroke in the cerebral infarction side with the expression of CX3CR1 in neurons.2. Longa count scores in each group for postoperative awake:group for 06 hours is 2±1.5; group for 12 hours is 2±0.5; group for 24 hours is 2±0.5; group for 48 hours is 2.75±0.75; group for 72 hours is 1.75±0.25,between group differences were no statistically significant; Longa count scores in each group for specimens were taken before:group for 06 hours is 0.5±0.5; group for 12 hours is 2±0.5; group for 24 hours is 2±0.5; group for 48 hours is 2.75±0.75; group for 72 hours is 3.25±0.25,between group differences were statistically significant.3、With the passage of time,the CX3CR1 positive cell counts in the infarct side: group for 06 hours is 382±67/mm2; group for 12 hours is 1138±64/mm2; group for 24 hours is 769±65/mm2; group for 48 hours is 323±53/mm2; group for 72 hours is 152±35/mm2,between group differences were statistically significant; the mean CX3CR1 positive cell counts in the cortex infarction side:group for 06 hours is 126±45/mm2; group for 12 hours is 274±31/mm2; group for 24 hours is 207±18/mm2; group for 48 hours is 54±18/mm2; group for 72 hours is 36±9/mm2,between group differences were statistically significant; the mean CX3CR1 positive cell counts in the hippocampal infarction side:group for 06 hours is 36±09/mm2; group for 12 hours is 126±09/mm2; group for 24 hours is 99±09/mm2; group for 48 hours is 40±04/mm2; group for 72 hours is 31±04/mm2,between group differences were statistically significant; the mean CX3CR1 positive cell counts in the striatum infarction side:group for 06 hours is 220±13/mm2; group for 12 hours is 445±13/mm2; group for 24 hours is 198±16/mm2; group for 48 hours is 81±09/mm2; group for 72 hours is 36±09/mm2,between group differences were statistically significant; the mean CX3CR1 positive cell counts in the ischemic penumbra side:group for 12 hours is 297±09/mm2; group for 24 hours is 216±18/mm2; group for 48 hours is 121±13/mm2; group for 72 hours is 31±04/mm2,between group differences were statistically significant; the mean CX3CR1 positive cell counts in the infarction area center side:group for 24 hours is 54±09/mm2; group for 48 hours is 27±09/mm2; group for 72 hours is 18±09/mm2,between group differences were no statistically significant; the mean CX3CR1 positive cell counts in the different parts of infarction side:group for the cortex infarction side is 140±24/mm2; group for the hippocampal infarction side is 66±07/mm2; group for the striatum infarction side is 196±12/mm2; group for the ischemic penumbra side is 166±11/mm2; group for the infarction area center side is 33±09/mm2,between group differences were statistically significant.4、With the passage of time,the average ratio of infarction area in each group (%) group for 06 hours is 15±02; group for 12 hours is 18±05; group for 24 hours is 38±04; group for 48 hours is 40±04; group for 72 hours is 48±03,between group differences were statistically significant.Conclusion1. The expression of CX3CR1 in both multiple infarction side of neurons.2. Expression CX3CR1 of neurons has occurrence after ischemic stroke of 06H, the counts of cells in 12H group was the highest, with the passage of time after the trend was gradually reduced.3.The expression of CX3CR1 in neuron cell counts in different regions is in the striatum expression quantity is the most, the infarction area center was the least.4. The counts of the expression CX3CR1 in positive neurons and Longa score and the infarct area variation trend is not consistent.
