Objective: By observing the influence of Qingreditanjiejing formula on lipids in serum, Ox-LDL, hs-CRP, ET-1 and Formula on electrocardiogram( ECG), on coronary artery spasm(CAS) in high-fat diet mice. To discuss the effects on the mice with hyperlipidemia and CAS and potential function points of the formula on the mouse CAS.Methods: After feeding the 140 C57BL/6J, SPF grade, 17-20 g weight mice one week with the normal fodder, divided into 7 group according to the weight stratified randomized block method. Normal control group, model control group, positive control Diltiazem hydrochloride group(HXS), positive control atorvastatin calcium tablet group(ATO), and QRDT low, middle, and high dose group, each group has 20 mice. Every group mice will be fed with high-fat fodders for 4 weeks except the normal control group(heat includes: Carbohydrates 20. 1%, fat 59. 8%, protein 20. 1%). The QRDT group will be fed with 5times, 10 times and 20 times dosage of clinical human one, namely gavages by 1.8, 3.6, 7.2g/kg granules; Then set up 2 positive control groups and feed the mice with diltiazem, atorvastatin calcium tablets according to the instruction to gavage with 0.040g/kg, 0.013g/kg dosage which are equivalent to 10 times of clinical dosage(4 mg/kg, 1.3 mg/kg); feed the normal control group and model control group with same volume solvent. Prepare corresponding concentration injectable Suspension with 0.5% sodium carboxymethyl cellulose solution before test and to gavage with 30ml/kg solution one time each today for 4 weeks. One hour after the last time dose, intraperitoneal injection with Pit( 60U/kg) to make modeling. Record the Ⅱ lead ECG and T wave displacement data respectively at 10 minutes before injection, and the minute after injection, 15 s, 30 s, 1, 3, 5, 10, 15 min after injection, take T wave displacement ≥50% as the judgment indicator. Test the content of TC, TG, HDL, and LDL, and use ELISA method to test the hs-CRP, ET-1, ox-LDL level.Results:(1) T wave displacement data: 1, 5, 10, 15 min after intraperitoneal injection Pit, T wave of the model group mice have obviously displacement compared with normal group(P<0.05), which shows CAS mice obviously myocardial ischemia and the modeling is successful. Compare with model group, the QRDT high, middle, low dosage group can resist the T wave displacement caused by Pit injection, the displacement range are exceeded 0.1m V(P<0.05) which have statistical significance. Compare with HXS, high and middle dosage group don’t have obvious T wave displacement(P>0.05) without statistical significance; T wave displacement data of the low dosage group is significantly increased, which have the statistical significance. There is no significant difference between the high, middle and low group, which don’t have any statistical significance.(2) lipids in serum: Compare with the normal group, the serum TC, TG, LDL-C content of model group mice have increased obviously(P<0.05), but the HDL-C content has decreased than the normal group(P<0.05), which shows the abnormal ipid metabolism with the coronary spasm mice. Compare with model group, the high dose group and ATO group can obviously decrease serum TC, TG, LDL-C content of the mice(P<0.05) and increase serum HDL-C content(P<0.05), The serum TC, TG, LDL-C, HDL-C level between the two groups have no significant difference(P>0.05).(3) ox-LDL: Compare with the normal group, the ox-LDL content of model group and all the dose groups has obviously increased(P<0.05), and the difference are with statistical significance; Compare with the model group, the ox-LDL content of high dose ATO group both decreased obviously(P<0.05), and the difference are with statistical significance; Compare with ATO group, middle and low dosage group and HXS group have significant changes(P<0.05), and the difference are with statistical significance; ox-LDL content of high dose group have no significant changes(P>0.05) with no statistical significance. Above all, high dose group and ATO group can effectively reduce the serum ox-LDL content of coronary spasm mice, and the two groups drug effect are mostly equal.(4) hs-CRP: Compare with the normal group, the hs-CRP content of model group and dose group have significantly increased(P<0.05), and the difference are with statistical significance; Compare with model group, the hs-CRP content of high, middle, and low dosage group and ATO group have significantly decreased(P<0.05), and the difference are with statistical significance; HXS group hs-CRP content are without significant changes(P>0.05) and no statistical significance; Compare with ATO group, high dosage and HXS group have big changes(P<0.05), and the difference are with statistical significance; there is no big changes in middle and low dosage group(P>0.05) with no statistical significance. Above all, QRDT high, middle and low dosage group can reduce the hs-CRP content and all dose group have obviously different, which have statistical significance.(5)ET-1: Compare with the normal group, ET-1 content of the model group and other dose groups has obviously increasing(P<0.01), and the difference has statistical significance; compare with the model group, ET-1 content of all the dose groups has obviously decreased(P<0.05), and the difference has statistical significance; Compare with HXS group, there is no significant different between ET-1 content of QRDT high, middle, low groups, no statistical significance; the ET-1 content of low dosage group and ATO group have clear changes(P<0.01) and the changes have statistical significance; there is no big difference between the high, middle and low group(P>0.05) with no statistical significance.Conclusion:(1) Qingreditanjiejing formula can improve ECG of coronary spasm mice, improve myocardial ischemia.(2) this formula can reduce the TC, TG, LDL-C content of the hyperlipidemia mice and adjust lipid metabolism disorders;(3) this formula can effectively reduce the Ox-LDL content of hyperlipidemia mice; which may be related to inhibiting the inflammatory response and anti lipid peroxidation;(4) this formula can effectively reduce the Ox-LDL content of CAS mice, inhibit the inflammatory response.(5)this formula can reduce the ET-1 content of coronary spasm mice, inhibit vasoconstriction, relieve coronary artery spasm.In conclusion: This formula can Effective regulation of lipid metabolism in mice with disorders,function of reduce the LDL-C content can direct lessen vascular endothelial damage; At the same time educe the Ox-LDL content inhibiting the inflammatory response and reducing the expression and secretion of ET-1. This formula can improve ECG of coronary spasm mice, Confirmed its function of ease cramps, treatment of myocardial ischemia Probably by reducing serum LDL-C effects achieved.
